Tissue level, activation and cellular localisation of TGF-β1 and association with survival in gastric cancer patients

Abstract: Transforming growth factor-b1 (TGF-b1), a tumour suppressing as well as tumour-promoting cytokine, is stored as an extracellular matrix-bound latent complex. We examined TGF-b1 activation and localisation of TGF-b1 activity in gastric cancer. Gastric tumours showed increased stromal and epithelial total TGF-b1 staining by immunohistochemistry. Active TGF-b1 was present in malignant epithelial cells, but most strongly in smooth muscle actin expressing fibroblasts. Normal gastric mucosa from the same patient sho… Show more

“…These findings were also associated with a poor prognosis for the patients. 45 Similar observations have been made in ovarian and colon carcinomas. 46,47 A strong local upregulation of the PA-system mediated through uPAR could in turn not only activate other proteases but also be important stimulators of cell proliferation and angiogenesis.…”

“…A-77 significantly inhibited the invasiveness of cancer cells with or without TGFh. Because gastric cancer cells produce TGFh1 (33,34), A-77 might decrease the autocrine manner of invasion ability for gastric cancer cells. The pathologic findings of the peritoneal nodules showed diffuse infiltration of cancer cells with the fibrous stroma on a control mouse.…”

“…To determine tissue localization of endoglin, immunohistochemistry was performed as previously described (23). Representative photomicrographs were taken with a Nikon Eclipse E900 microscope equipped with a Nikon DXM1200 digital camera.…”
