Tissue distribution profiles of multiple major bioactive components in rats after intravenous administration of Xuebijing injection by UHPLC–Q–Orbitrap HRMS

Abstract: Xuebijing injection (XBJI) is a traditional Chinese medicine prescription extracted from five Chinese herbs. Hydroxysafflor yellow A, oxypaeoniflorin, ferulic acid and benzoylpaeoniflorin are the main bioactive ingredients of XBJI. This paper presents an application of ultra-high-performance liquid chromatography-Q-exactive hybrid quadrupole-Orbitrap high-resolution mass spectrometry (UHPLC-Q-Orbitrap HRMS) to quantify four compounds of XBJI in rats various tissues for tissue distribution studies. The analytes… Show more

“…Like artemisinin used for malaria treatment, XBJ used as an injection is also a medicinal ingredient extracted from a traditional Chinese medicine. Its main components include hydroxysa or yellow A, ligustrazine, salvianic acid A, ferulic acid, paeoni orin, and protocatechuic aldehyde [23]. XBJ can effectively improve the prognosis of patients with paraquat poisoning, and the potential mechanisms include the following.First, by inhibiting the overexpression of proin ammatory factors, such as tumor necrosis factor α (TNF-α), platelet-derived growth factor (PDGF), insulin-like growth factor-1 (IGF-1),transforming growth factor-β1 (TGF-β1), interleukin (IL)-1, and IL-6; blocking the TGF-β1/ERK signaling pathway [24]; promoting the expression of anti-in ammatory factors, such as IL-10; re-establishing the balance of in ammatory/anti-in ammatory transmitters; attenuating in ammation; and exerting organ protection.…”

“…Among these compounds, senkyunolide I-7-O-â-glucuronide, 3-O-methyltanshinol, protocatechuic acid, and 3-O-methylsalvianolic acid B are the in vivo metabolites of senkyunolide I, tanshinol, protocatechuic aldehyde, and salvianolic acid B, respectively; the unchanged compound protocatechuic aldehyde could not be detected in human plasma samples [29,[49][50][51]. Several other research groups also measured circulating herbal compounds in their unchanged forms in rats receiving XueBiJing based on developed bioanalytical assays [52][53][54][55]. Zuo et al investigated the tissue distributions of several bioactive compounds in rats after they intravenously received XBJ, and the levels of exposure to four compounds (i.e., hydroxysafflor yellow A, paeoniflorin, ferulic acid, and benzoylpaeoniflorin) were found to be high in the kidneys, liver, stomach, and intestines [52].…”

“…Diels (Danggui). Ultra-high-performance liquid chromatography coupled with quadrupole Orbitrap high-resolution mass spectrometry (UHPLC-Q-Orbitrap HRMS) indicates that the primary components of XBJ are hydroxysafflor yellow A, oxypaeoniforin, senkyunolide I, and benzoylpaeoniflorin [ 77 ]. XBJ has been approved for use in the treatment of severe infections (sepsis) in critically ill patients (China Food and Drug Administration; Beijing, China, Number Z20040033).…”
