Tissue distribution of hepatopoietin-A: a heparin-binding polypeptide growth factor for hepatocytes.

Abstract: Hepatopoietin-A (HPTA) is a heparinbinding polypeptide growth factor which consists of a heavy and a light polypeptide chain with molecular weights of 70,000 and 35,000, respectively. It stimulates DNA synthesis in primary cultures of normal rat hepatocytes in serum-free medium. The complete purification and characterization of HPTA from rabbit serum were reported by us elsewhere. Recently we have determined the amino-terminal amino acid sequence of the rabbit HPTA light chain up to 24 residues and have shown … Show more

“…These in vivo results extend previous in vitro findings suggesting that HGF is one of the most potent mitogens and insulinotropic agents for beta cells (14,19,20). Since HGF is produced in the pancreas (12)(13)(14)(15)(16)(17) and the c-Met receptor is localized in the islet beta cells (14), it seems likely that HGF may play a paracrine and/or autocrine physiological role in the growth and development of the pancreatic islet.…”

“…HGF has been detected immunohistochemically in the exocrine portion of rabbit pancreas, and in rat and human pancreatic islet cells (15)(16)(17). Tissue-type plasminogen activator has been detected in the rat endocrine pancreas, preferentially in somatostatin cells (18).…”

“…The demonstration that HGF also had a strong effect in Eck's fistula model is the first evidence of its in vivo potency. HGF is produced in multiple sites, including the exocrine portion of the pancreas (46), and its physiological role remains to be determined. However, using in vitro testing for growth effects, researchers might pass over as unexciting not only insulin and IGF-II but also cyclosporine (47), FK 506 (47) and HSS.…”

“…Expressão elevada do met/receptor do HFG nas células foliculares neoplásicas parece dever-se a uma relação parácrina entre o fator estimulante e o receptor já que o HGF é secretado pelas células parafoliculares tireoidianas 25,26 .…”
