Tissue‐based quantitative proteomics to screen and identify the potential biomarkers for early recurrence/metastasis of esophageal squamous cell carcinoma

Abstract: Esophageal squamous cell carcinoma (ESCC) is the eighth cause of cancer‐related deaths worldwide. To screen potential biomarkers associated with early recurrence/metastasis (R/M) of ESCC patients after radical resection, ESCC patients were analyzed by a comparative proteomics analysis using iTRAQ with RPLC‐MS to screen differential proteins among R/M groups and adjacent normal tissues. The proteins were identified by qRT‐PCR, Western blotting, and tissue microarray. The protein and mRNA expression difference o… Show more

“…PHB2 is a multifunctional protein involved in numerous vital cellular processes including signal transduction, gene transcription, cell survival, metabolism, inflamemation, and apoptosis 8 , 9 . Increasing evidence shows that PHB2 is highly expressed in prostate cancer 10 , liver cancer 11 , esophageal squamous cell carcinoma 12 , and diffuse large B cell lymphomas 13 compared with normal tissues. Moreover, a correlation between PHB2 expression and patients' clinicopathologic characteristics has been observed, and PHB2 is regarded as an independent prognostic marker for these cancers 10 - 13 .…”

“…In recent years, many studies have comprehensively interrogated the proteome of EC in search for biomarkers to aid disease management at different levels, including diagnosis, patient stratification, prognosis and responses to treatment [5][6][7]. Despite promising, conventional proteomics approaches on EC are yet to deliver targetable molecular signatures due to insufficient cancer specificity.…”

“…Surgical resection is recognized as the preferred treatment for patients with newly diagnosed ESCC. However, high rates of tumor recurrence are notable (4,5). Neoadjuvant chemotherapy or chemoradiotherapy have been demonstrated to prolong overall survival for patients with ESCC (6)(7)(8).…”
