Tip enhanced Raman spectroscopy imaging of opaque samples in organic liquid

Abstract: Implementation of Tip Enhanced Raman Spectroscopy in liquid is still a challenge. We demonstrate herein its feasibility in an upright illumination/collection configuration. Through a thin layer of organic solvent covering the sample, laser focussing on the tip is possible, enabling TERS imaging in liquid.

“…TERS in liquid environments was first demonstrated in 2009 using silver-coated AFM probes, with self-assembled molecular monolayers used to protect the tips from contamination [130]. Further applications of liquid TERS have since been demonstrated, including the analysis of lipid bilayers [131], the adsorption of probe molecules on Au(111) [132], and TERS imaging in organic liquids [133]. It has taken several years since the first demonstration of liquid TERS to develop EC-TERS but 2015 saw major advances from two independent groups [134,135].…”

“…In addition, STM-TERS can also be manipulated in liquid. A few studies have been carried out by electrochemical STM-TERS (EC-TERS) [ 53 , 54 , 55 , 56 ], where the STM probe was immersed in some suitable solvent for the electrochemical measurement. In EC-TERS, a suitable liquid is primarily required to maintain the tunneling current, and meanwhile a proper coating on the tip should be satisfied to ensure that the Faradaic current doesn’t interfere with the tunneling current.…”

“…Emerging applications of TERS using a liquid medium [180][181][182] and improvements in the data acquisition speed by Van Duyne et al 183 will make this technique a promising tool for imaging structural and dynamic events in live cell membranes. Therefore the sample preparation is easier, and the potential of altering the membrane structure and dynamics during the imaging process is limited.…”
