TIMP-1 gene polymorphism: are genetics able to predict outcome of septic patients?

Abstract: The multicenter study conducted by Lorente and coworkers - published in the previous issue of Critical Care - suggests that levels of tissue inhibitor of metalloproteinase (TIMP)-1 in association with the 372 T/C genetic polymorphism of TIMP-1 are promising markers to predict the clinical outcome of septic patients. TIMPs bind to active matrix metalloproteinases and, amongst other effects, inhibit their proteolytic activity of the extracellular matrix. Previous clinical studies showed increased plasma levels o… Show more

“…Patients with T allele show higher serum levels of TIMP‐1 19 . The association between this TIMP‐1 polymorphism and other diseases was reported in previous studies, for example, systemic sclerosis, 20 sepsis, 21 chronic obstructive pulmonary disease, 22 and primary open‐angle glaucoma 23 . However, there is scarcity in the studies investigating the association between this polymorphism and dermatological disorders.…”

“…Gene polymorphism refers to the variation of gene sequences in the same population. Gene polymorphisms determine the susceptibility or tolerance of the body to stress stimuli and the diversity of clinical manifestations and drug therapeutic effects, thus further affecting an infected host's gene expression and prognosis (Behnes et al, 2013;Thompson et al, 2014). With the deepening understanding of genomics, genetic differences are considered to be the internal basis for the occurrence and development of many diseases.…”

“…In the previous issue of Critical Care , Behnes and colleagues [ 1 ] provide an interesting commentary on our study showing that septic patients with the T-allele in 372 T/C (rs4898) genetic polymorphism of the tissue inhibitor of metalloproteinase-1 (TIMP-1) had higher mortality and higher TIMP-1 serum levels than those without it [ 2 ].…”
