Time-to-Onset Analysis of Drug-Induced Long QT Syndrome Based on a Spontaneous Reporting System for Adverse Drug Events

Abstract: Long QT syndrome (LQTS) is a disorder of the heart’s electrical activity that infrequently causes severe ventricular arrhythmias such as a type of ventricular tachycardia called torsade de pointes (TdP) and ventricular fibrillation, which can be fatal. There have been no previous reports on the time-to-onset for LQTS based on data from spontaneous reporting systems. The aim of this study was to assess the time-to-onset of LQTS according to drug treatment. We analyzed the association between 113 drugs in 37 the… Show more

“…SRS is one of the primary tools used in pharmacovigilance because they reflect the realities of clinical practice (Poluzzi et al, 2012). With larger numbers of faithful reports, the FAERS database should help for optimizing pharmacotherapy (Sasaoka et al, 2016). Furthermore, Concato et al reported that the results of well-designed observational studies do not systematically overestimate the magnitude of the effects of treatment as compared with those in randomized, controlled trials on the same topic (Concato, Shah, & Horwitz, 2000).…”

“…For our resident's medications, the peak occurrence is expected to be about 2 weeks following drug initiation; however, time to onset ranges widely from 2 to 30 days. Thus if ECG monitoring is implemented, it should extend for 2 to 4 weeks …”

“…Thus if ECG monitoring is implemented, it should extend for 2 to 4 weeks. 53 Table 3 outlines recommendations for issues unresolved in this population. In the absence of evidence in older adults, healthcare professionals remain accountable for optimizing drug therapy outcomes and minimizing the risk of adverse drug events.…”

“…If β is greater than 1 and the 95% CI of β excludes the value of 1, the hazard is considered to increase over time. [17][18][19][20]24,25) We investigated the relation between the number of reports and seasonality of DIP. We analyzed the onset day of AEs and then obtained the number of AE reports per month between April 2004 and November 2016 from REAC table of JADER.…”
