Time-resolved immunofluorometric assay of 34-kDa somatomedin-binding protein.

Abstract: In this time-resolved immunofluorometric assay for the 34-kDa somatomedin-binding protein (SmBP), affinity-purified polyclonal antibodies are used, along with solid-phase separation of bound and free analyte. The first antibody is bound to polystyrene microtiter wells; the second is labeled with europium(III) chelate. The detection limit of the method is 0.25 microgram/L, much lower than that (about 8 micrograms/L) for radioimmunoassay. By immunofluorometric assay, SmBP is detectable, and could be accurately q… Show more

“…Insulin was measured by radioimmunoassay using a polyclonal antiserum (MF/GP/9) from Guildhay Ltd, Guildford, Surrey and IGF-I by RIA after acid-ethanol extraction of the serum using reagents from Amersham International, Amersham, Bucks (Daughaday et al, 1980). The 34kDa IGF-BP (IGF-BP-I, PP 12) was measured by an immunofluorometric assay as described by Koistinen et al (1987).…”

“…Blood samples were obtained for analysis of the serum concentrations of IGFBP‐1 and PP14 by immunofluorometric assays 4,5 with an inter‐assay coefficient of variation of less than 10%. The sensitivity of the PP14 assay, as used in the present study, was 3 μg/1.…”

“…The IGFBP-1 concentration was measured by a specific monoclonal time-resolved immunofluorometric assay performed essentially as described earlier (Koistinen et al, 1987;Angervo et a/., 1992). The sensitivity of the assay was 0.1 pg/l.…”
