Thyroid Transcription Factor 1 Is Calcium Modulated and Coordinately Regulates Genes Involved in Calcium Homeostasis in C Cells

Abstract: Thyroid transcription factor 1 (TTF-1) was identified for its critical role in thyroid-specific gene expression; its level in the thyroid is regulated by thyrotropin-increased cyclic AMP levels. TTF-1 was subsequently found in lung tissue, where it regulates surfactant expression, and in certain neural tissues, where its function is unknown. Ligands or signals regulating TTF-1 levels in lung or neural tissue are unknown. We recently identified TTF-1 in rat parafollicular C cells and parathyroid cells. In this … Show more

“…Cells were grown in Coon's modified F-12 medium (Sigma Chemical Co) containing 5% heat-treated, mycoplasma-free calf serum (GIBCO Laboratories Life Technologies), 1 mM nonessential amino acids (GIBCO), and supplemented with six hormones or growth factors (termed 6H) that included bovine TSH (1 10 10 M), insulin (10 µg/ml), cortisol (0·4 ng/ml), transferrin (5 µg/ml), glycyl--histidyl--lysine acetate (10 ng/ml), and somatostatin (10 ng/ml). Bovine Tg was added directly to the culture medium as described previously (Royaux et al 2000, Suzuki et al 1998c, 1999c.…”

“…surfactant proteins and the Clara cell secretory protein) genes (2-7). TTF-1 has been reported to be expressed also in the adult rat parathyroid cells and several other adult tissues (including skin, esophagus, retina, anterior pituitary, cerebellum, and hippocampus) (8,9). Recently, it has been shown that postnatal hypothalamic TTF-1 expression is strongly and transiently up-regulated immediately before puberty in female rats and is associated with the neuroendocrine process of female sexual development (10).…”

“…The column was equilibrated in buffer A (30 mM HEPES, 0.1 mM EGTA, 0.5 mM DTT, 0.5 mM PMSF) þ 80 mM NaCl and loaded with the nuclear extract prepared from HeLa cells overexpressing Pax 8. The nuclear extract, prepared according to Suzuki et al (1998), was diluted with buffer A to decrease the salt concentration to 80 mM. The column was washed with buffer A þ 80 mM NaCl and eluted with buffer A þ 1 M NaCl.…”
