Thyroid hormone modulation of the hypothalamic growth hormone (GH)-releasing factor-pituitary GH axis in the rat.

Abstract: Both thyroid hormone and hypothalamic growth hormone (GH)-releasing factor (GRF) facilitate pituitary somatotroph function. However, the pathophysiological role of thyroid hormone in GRF secretion is less well understood. Thyrotoxicosis, induced by administration of thyroxine (T4) in rats, inhibited both pituitary GH levels and immunoreactive GRF secretion from incubated hypothalamus. At the highest dose of T4 given for 12 d, GRF secretion and pituitary GH decreased by 50 and 39%, respectively. Hypothyroidism … Show more

“…The secretion of m G R F in response to 50 mM K + was highly constant with mean (+SE) values ranging from 13.2iO.2 to 16.2+2.1 pg/hypothalamus/30 min in 6 experiments, each using 10-12 animals. This value is only one third of the GRFreleasing capacity of the rat hypothalamus, which we observed previously by identical experimental methods (36). No definite answer is available as to what determines the species difference in the releasing capacity of G R F between the two species.…”

“…We previously reported the hypothalamic content and in vitro secretion of G R F in the rat (35,36). It would be interesting to refer to the species difference in these parameters on GRF between rat and mouse.…”

“…The intensity of GRF receptor mRNA was normalized by that of f3-actin mRNA. Hypothalamic incubation was performed as described [6,7], using 12 male and 12 female rats in each of three separate experiments.…”
