Thyroid Hormone Activation in Vascular Smooth Muscle Cells Is Negatively Regulated by Glucocorticoid

Abstract: Background: Type 2 iodothyronine deiodinase (D2) catalyzes the production of triiodothyronine from thyroxine. D2 is present in rat aorta media, and there is a circadian variation in the D2 expression. In rat aorta media, the D2 activity exhibited the maximal value at 1200 hour and low value between 1800 and 2400 hour. To understand the mechanisms that induce the circadian variation in the D2 expression, we examined the effects of glucocorticoid on the D2 activity and mRNA in rat aorta media and cultured vascul… Show more

“…Thyroid hormones has a known role in BAT, specially triiodothyronine (T3) which acts promoting thermogenesis and its local activity is mediated by type 2 iodothyronine deiodinase (Dio2) that converts intracellular thyroxine (T4) to T3 [50]. Studies show that hypercorticosteronemia appears to negatively regulate Dio2 activity in peripheral tissues, including BAT [51,52]. Thus, the hypercorticosteronemia observed in SL rats may lead to a reduction in Dio2 and T3, contributing to IBAT hypoactivity…”

“…Presence of iodothyronine deiodinase enzymes in SMCs of the aorta in rats (Toyoda et al 2009) and the expression of TH receptors in coronary ECs of mice (Makino et al 2009) and bovine aorta (Hu et al 1994) suggests that during adulthood, arteries are a target for THs. They bind to integrin αvβ3 in the plasma membrane of the ECs (Bergh et al 2005) and activate the mitogen-activated protein kinases cascade, increasing angiogenesis (Davis et al 2004) and proliferation of ECs via the fibroblast growth factor (Al Husseini et al 2013).…”

“…Serum concentrations of T 3 were measured using an electrochemiluminescence immunoassay kit (Roche Molecular Biochemical, Indianapolis, IN, USA). This methodology has been frequently used in the assessment of total serum levels of T 3 .…”
