Thymoquinone inhibits cell proliferation, migration, and invasion by regulating the elongation factor 2 kinase (eEF-2K) signaling axis in triple-negative breast cancer

Abstract: Our study provides first evidence that TQ treatment inhibits cell proliferation, migration/invasion, and tumor growth, in part through the inhibition of eEF-2K signaling in TNBC. Thus, our findings suggest that systemic TQ treatment may be used as a targeted therapeutic strategy for the inhibition of eEF-2K in TNBC tumor growth and progression.

“…Cell survival depends on the balance between cell proliferation and apoptosis. TQ has been found to inhibit the survival of many cancer cells derived from breast cancers [ 65 , 67 , 77 , 83 ], pancreatic cancer [ 84 ], gastric cancer [ 85 ], primary effusion lymphoma [ 43 ], prostate cancer [ 86 , 87 ], kidney cancer [ 88 ], squamous cell carcinoma [ 89 ], renal cell carcinoma [ 90 ], and cholangiocarcinoma [ 63 ], primarily through attenuation of the activity of AKT, a crucial kinase associated with cell survival. TQ inhibits the activation of AKT through ROS-dependent mechanisms and/or through activation of the lipid phosphatase, phosphatase and tensin homolog (PTEN).…”

“…In cancer, it is deemed therapeutic through its interference with cell proliferation, apoptosis, cell invasion, and metastasis in cancer cells using different in vitro and in vivo models. [6][7][8][9] Theses anticancer effects were reported in various types of cancers such as breast cancer cells, 10 colon cancer, 11 lung cancer, 12 cervical squamous carcinoma, 13 ovarian cancer, 14 prostate cancer cell, 15 and osteosarcoma. 16 In addition, synergistic therapeutic effects were elicited when TQ was given in combination with other chemotherapeutic agents.…”

“…Black cumin seed ( Nigella sativa ) and thymoquinone exercised a protective effect as they reduced the breast carcinogens and decreased the gene expressions of Id-1, Brac2, Bra1 and p53 mutations [ 249 ]. In another study, in vivo administration of thymoquinone (20 and 100 mg/kg) significantly decreased the progression of MDA-MB-231 tumors and inhibited the eukaryotic elongation factor 2 kinase (eEF-2K) in an orthotopic tumor mouse model of triple-negative breast cancer [ 250 ].…”
