Thymic stromal lymphopoietin is released by human epithelial cells in response to microbes, trauma, or inflammation and potently activates mast cells

Abstract: Compelling evidence suggests that the epithelial cell–derived cytokine thymic stromal lymphopoietin (TSLP) may initiate asthma or atopic dermatitis through a dendritic cell–mediated T helper (Th)2 response. Here, we describe how TSLP might initiate and aggravate allergic inflammation in the absence of T lymphocytes and immunoglobulin E antibodies via the innate immune system. We show that TSLP, synergistically with interleukin 1 and tumor necrosis factor, stimulates the production of high levels of Th2 cytokin… Show more

“…This is consistent with recent in vitro studies, which suggest that TSLP expression in epithelial cells requires proinflammatory cytokines such as TNF-a [11][12][13]. Therefore, it is possible that mast cell-derived inflammatory cytokines including TNF-a generated at the site of allergic rhinitis may play an important role for the induction of TSLP expression in the nasal epithelium.…”

“…Conversely, TSLP has been shown to stimulate the production of high levels of Th2 cytokines by human mast cells synergistically with IL-1 and TNF-a [12]. Therefore, it is possible that there is an important interaction between epithelial cells and mast cells via TSLP and proinflammatory cytokines for the development of allergic rhinitis.…”

“…TSLP can stimulate mast cells and dendritic cells to initiate the innate allergic immune responses. 95 In the latter phases, TSLP activated DCs mediate the T cell dependent adaptive phase of the allergic immune response.…”
