Thymic Stromal Lymphopoietin Is a Key Mediator of Breast Cancer Progression

Abstract: Inflammation is a double-edged sword that can promote or suppress cancer progression. Here we report that thymic stromal lymphopoietin (TSLP), an IL-7-like type 1 inflammatory cytokine that is often associated with the induction of Th2-type allergic responses in the lungs, is also expressed in human and murine cancers. Our studies with murine cancer cells indicate that TSLP plays an essential role in cancer escape, as its inactivation in cancer cells alone was sufficient to almost completely abrogate cancer pr… Show more

“…The suppression of inflammation with the corticosteroid budesonide decreased CD4 + T cell recruitment and metastasis to the lung. Another study concurred with these findings in that activated CD4 + T cells from thymic stromal lymphopoietin-induced airway allergic inflammation resulted in increased tumor growth and lung metastasis [42]. It is well established that macrophages are one of the leukocyte subpopulations that contribute to enhanced tumor growth and metastasis.…”

“…Unlike observations seen by this study, our observations may have occurred due to the preexisting Th-2 skewed phenotype associated with the 4T1 tumor model. 45 This phenotype may have exacerbated the anaphylactic response to foreign antibodies. Additionally, it has been shown that both Th1 and Th2 responses can stimulate IgG1 production and that a switch from Th2 to Th1 can occur and suppress IgE responses; this can also occur in an organ specific manner if mixed Th1/Th2 responses occur.…”

“…8,[10][11][12] The hypothesis of an implication of TSLP in the tumor microenvironment stemmed from earlier studies in breast and pancreatic cancer, both in humans and in mice. [13][14][15][16] TSLP expression was associated in these studies with the presence of a Th2-biased inflammatory response, characterized by the expression in T cells of the transcription factor GATA-3, and the production of IL-4, IL-5, IL-13, and TNF. Because a Th2 response has been associated with a poor prognosis, 17,18 TSLP was proposed to be associated with tumor progression and to indicate a bad prognosis of the tumor.…”
