THSD7A expression in human cancer

Abstract: We recently described a case of a Thrombospondin Type-1 Domain containing 7A (THSD7A) associated membranous nephropathy in a female patient who was synchronously suffering from a THSD7A-positive malignancy. We here investigated the role of THSD7A as a new potential tumor antigen by evaluating over 20 000 tissue spots in more than 70 different tumor entities by immunohistochemistry using tissue microarrays. THSD7A expression was highly variable in different neoplasias with differing staining patterns. Both gain… Show more

“…The second patient had previously been treated for renal cell carcinoma as well as breast carcinoma. Many different types of cancer have been shown to have some degree of positive immunohistochemical staining for THSD7A 6 and a previous case report makes a strong argument for the connection between THSD7A-associated membranous nephropathy and malignancy. 8 In this report, the patient’s tumor was found to be positive for THSD7A on immunohistochemical analysis and have increased copy number of the THSD7A gene.…”

“… 5 Recent reports have implicated malignancy as the underlying etiology in some cases of THSD7A-associated membranous nephropathy. 6 , 7 , 8 In an effort to further define the clinicopathologic spectrum of THSD7A-associated membranous nephropathy, we undertook the current retrospective study of 31 patients with follow-up of up to 2 years. We also sought to establish the utility of THSD7A staining in renal biopsies and its correlation with antibody titers for the detection of THSD7A-associated membranous nephropathy.…”

“…In the glomeruli, granular staining for THSD7A along the GCW was observed with negative staining for PLA2R. In the rectal cancer tissue, THSD7A staining was positive, showing a mainly luminal pattern, which was reported as the staining pattern of rectal cancer [12]. THSD7A staining of metastatic lymph nodes revealed deposition in the secondary lymph follicles and in the sinus.…”

“…A recent study catalogs THSD7A expression by a variety of tumors, and finds strong THSD7A staining in prostate, breast, renal, and colorectal carcinomas. 76 However, overexpression or loss of THSD7A expression was not consistent; in colorectal and renal cell carcinoma, a lack of THSD7A was associated with worse outcome. In contrast, in prostate cancer, overexpression of THSD7A was associated with worse outcomes.…”
