Thrombomodulin distribution during murine development

Abstract: Thrombomodulin is a transmembrane glycoprotein involved in the regulation of clot formation. Previous work has shown that in the adult mouse, thrombomodulin expression is primarily located on the luminal surface of endothelial cells; whereas during development, thrombomodulin has been localized to the placental parietal endoderm, the neuroepithelium, head mesenchyme, lung bud, and atrium of Day 10.5 post coitum embryos. We examined the expression of thrombomodulin in the adult mouse and throughout murine devel… Show more

“…By this hypothesis the absence of Thrombomodulin from blood vessel endothelium may cause excessive activation of the embryonic blood coagulation system [17]. We cannot state at present the exact meaning of a reduced expression of Thrombomodulin in placental tissue.…”

“…Abolishing Tie2 function by knocking out the receptor (3,13) or its recently identified activating ligand (2,16) results in embryonic lethality in mice between embryonic days E9. 5 and E10.5 (3,13,16). This knowledge indicates the importance of understanding the mechanisms of Tie2 gene regulation during (patho)physiological processes.…”

“…compound (Allegiance Health Care Corp., Omaha, NE). Formalin-fixed, paraffin-embedded sections of all wounds were stained with Masson's trichrome or analyzed for murine thrombomodulin by immunoperoxidase staining, using the monoclonal antibody 273-34A 21 (generously provided by Dr. Stephen J. Kennel, Oak Ridge National Laboratory, Oak Ridge, TN) as described previously. 14 To detect ␤-galactosidase activity, frozen sections were fixed for 5 minutes in acetone and washed two times with phosphate-buffered saline.…”
