Thrombin specificity

Abstract: Selective cleavage of polypeptides by a-thrombin can be reasonably predicted [Chang, J.-Y. (1985) Eur. J . Biochem. 151,2241. This knowledge was applied to the selective cleavage of antibody light chains with the aim of producing intact fragments of both variable region and constant region. (a) Mouse K light chains 10K26 and 10K44 from anti-(azobenzene arsonate) antibodies contain 20 Arg/Lys-Xaa bonds. Only two of them, one ProArgThr bond located at the joint of the variable region with the joining peptide and… Show more

“…2) and, as stated above, thrombin's long-known requirement for arginine in position P1 was reproduced [8], [9], [12], [35]. In position P2, the well-established proline [7], [8], [11], [12], [36], [37] dominated (71%), but also aliphatic aa were tolerated. P2 glycine, valine or isoleucine were together present in 23% of the sequences.…”

“…Thrombin is a protease that recognizes LVPR^GS and cleaves between arginine and glycine [190][191][192]. Although thrombin cleavage at the designated sequence is relatively specific, there have been reports of thrombin cleavage at alternative sites [193].…”
