Thrombin Augments Vascular Cell-dependent Migration of Human Mast Cells: Role of MGF

Abstract: SummaryRecent data suggest that auricular thrombosis is associated with accumulation of mast cells (MC) in the upper endocardium (where usually no MC reside) and local expression of MGF (mast cell growth factor) (25). In this study, the role of vascular cells, thrombin-activation and MGF, in MC-migration was analyzed. For this purpose, cultured human auricular endocardial cells (HAUEC), umbilical vein endothelial cells (HUVEC) and uterine-(HUTMEC) and skin-derived (HSMEC) microvascular endothelial cells were e… Show more

“…Consistent with previous data (28), rHuSCF induced migration of HMC-1 in a dose-dependent manner ( Figure 5B). An antibody against SCF was found to block both SFB supernatant-induced ( Figure 5A) and SCF-induced ( Figure 5C) migration of HMC-1, but did not inhibit rHuC5a-induced HMC-1 migration (results not shown).…”

“…Histamine, proteases, TNF and PGs are known to induce vasodilation and increase vascular permeability [68]. Endothelial cells release IL-3 and thrombin, which enhance migration, proliferation and local differentiation of mast cells [69]. Mast cells increase fivefold along wound edges, an event caused by chemokine monocyte chemoattractant protein-1 and transforming growth factor (TGF)-β, a vital wound healing cytokine, which is also a potent chemoattractant for mast cells [70].…”

“…PAR-1 has been shown to be active in the release of IL-6 from murine mast cells (13) and NO from rat peritoneal mast cells (14). Thrombin may also affect mast cell biology through the release of mast cell chemotactic factors from endothelial cells (15).…”
