Three N-terminal Variants of the AE2 Cl-/HCO3- Exchanger Are Encoded by mRNAs Transcribed from Alternative Promoters

Abstract: Multiple AE2 Cl ؊ /HCO 3 ؊ exchanger mRNAs have been identified in rat. To determine the genetic basis for these mRNAs and whether they encode different variants of the exchanger, we used both rapid amplification of cDNA ends and S1 nuclease protection protocols and examined the organization of the gene. mRNAs encoding three N-terminal variants of AE2 (AE2a, AE2b, and AE2c) were identified and shown to be transcribed from alternative promoters. The AE2a transcription unit consists of 23 exons, with exons 1 and… Show more

“…Our recent data on HNF1a being able to transactivate alternate expression [17] strengthen the view that alternative AE2 isoforms might be related to the possibility of liver cells increasing their expression in response to certain stimuli for bicarbonate secretion. Noticeably, the HNF1 consensus sequence found in the human alternate overlapping promoter within intron 2 is also observed in rat [7] and mouse [33] orthologs. Moreover, both alternative messages AE2b 1 and AE2b 2 are expressed in the liver in these three species, the rat included.…”

“…Moreover, both alternative messages AE2b 1 and AE2b 2 are expressed in the liver in these three species, the rat included. Thus, using RT-PCR we could not only confirm the rat liver expression of AE2b 1 mRNA (previously referred to as just AE2b mRNA [7]) but also ascertain the liver expression of AE2b 2 mRNA (not shown).…”

“…An adenoviral vector was designed for the intracellular expression of small hairpin RNA (shRNA-1) toward this C-terminal sequence, which is common to all rat and human N-terminal AE2 variants. 27,28 As a negative control, we used an adenoviral Fig. 1.…”

“…The secretion of ϳ160 mM HCl and ϳ17 mM KCl by gastric parietal cells requires the coordinated activities of ion transporters on both apical and basolateral membranes. There is strong evidence that the major mechanism for Cl Ϫ uptake and HCO 3 Ϫ extrusion across the basolateral membrane is Cl Ϫ / HCO 3 Ϫ exchange (38 -42), which probably involves one or more variants of AE2 (31,43 stomachs, whereas the Na ϩ ,K ϩ -ATPase mediates K ϩ uptake and Na ϩ extrusion. Given the copious quantities of fluid secreted by the parietal cell, it is critical that cellular volume homeostasis be maintained.…”
