Three-Dimensional Visualization of Ultraviolet-Induced DNA Damage and Its Repair in Human Cell Nuclei

Abstract: The two major forms of DNA damage produced by 254 nm UV light are cyclobutane pyrimidine dimer (CPD) and (6-4) photoproduct (6-4PP). Both photolesions are repaired in normal human cells by nucleotide excision repair; however, little is known about where CPD or 6-4PP are repaired in relation to the various subnuclear structures. This study aimed to produce a three-dimensional demonstration of UV-induced DNA damage and its repair in human cell nuclei. We first investigated the repair kinetics of CPD and 6-4PP us… Show more

“…This procedure was described previously. [46][47] Briefly, 2 x 10 4 cells seeded/well in 24 well culture plates were infected with Ad-LacZ or Ad-AS-BRCA1 at 40 MOI for 24 hrs, then irradiated with 20 J/m 2 UVC. Cells were fixed immediately, at 6 hrs, 24 hrs, and 48 hrs after irradiation with cold (4°C) methanol-acetic acid (3:1) for one hour.…”

“…This procedure was described previously (26,27). Briefly, 5 ϫ 10 6 cells were seeded on 15-cm culture dishes and irradiated with 0, 15, 22.5, and 30 J͞m 2 UVC as described above.…”

“…Despite these detrimental effects of UVR, it is required to produce vitamin D 3 in skin cells. Vitamin D 3 is produced in the skin following exposure of its precursor, 7-dehydrocholesterol, to UVR (6). Following 2 successive hydroxylation reactions, the biologically active steroid hormone 1a,25-dihydroxyvitamin D 3 [1,25(OH) 2 D 3 ] is formed.…”
