Three‐dimensional localisation of NANOG, OCT4, and E‐cadherin in porcine pre‐ and peri‐implantation embryos

Abstract: The expression patterns of NANOG and OCT4 have previously been reported to differ markedly between mammalian species indicating distinct species-specific roles during development. We investigate the three-dimensional expression pattern of NANOG and OCT4 in porcine pre-and peri-implantation embryos. The expression of NANOG differed remarkably from that reported in other species. NANOG was not detected in the inner cell mass of hatched porcine blastocysts, but later appeared in the epiblast and hypoblast of sphe… Show more

“…In contrast to the pluripotency markers, the lineage porcine blastocysts (Day 9-10 development). 48 In the present study, we were able to detect NANOG in both the porcine epiblast and hypoblast and the mRNA transcript levels were higher in the hypoblast than that in the epiblast. This finding is in contradiction with known expression of NANOG in the mouse blastocyst.…”

“…After all, there exists a long spanning blastocyst stage and different developmental process in porcine embryos. And a recent study also demonstrated that Nanog was detected later in the epiblast and hypoblast of spherical blastocysts of pig (Wolf et al, 2011). But in early developmental stages before morula, it must play other regulatory roles different with that in the later stage, which is still needed to be identified.…”

“…In the pig however, primitive endoderm appears to have already formed before NANOG is expressed. Recently, Wolf et al (2011) found what appears to be a sequential expression of OCT4 and NANOG in the pig. OCT4 but not NANOG appears initially in the ICM and is followed later by co-localised expression of both of these genes in the epiblast; with subsequent downregulation of NANOG by the time the primitive streak develops.…”
