Three-dimensional Culture Regulates Raf-1 Expression to Modulate Fibronectin Matrix Assembly

Abstract: Oncogenic transformation has been associated with decreased fibronectin (FN) matrix assembly. For example, both the HT-1080 fibrosarcoma and MAT-LyLu cell lines fail to assemble a FN matrix when grown in monolayer culture (2-dimensional [2D] system). In this study, we show that these cells regain the ability to assemble a FN matrix when they are grown as aggregates (3-dimensional [3D] system). FN matrix assembly in 3D correlates with decreased Raf-1 protein expression compared with cells grown in monolayer cul… Show more

“…Further, expression of α-SMA in normal ovarian fibroblast has been previously reported in some parts of the normal ovary [46]. Separately, we and others have shown that while total levels of FAK expression remain unchanged in fibroblastic 2-D vs. 3-D cultures, the levels of tyrosine phosphorylated (activated) pFAKY 397 is greater in 2-D culture conditions [43,[47][48][49][50][51][52][53][54][55]. The housekeeping protein glyceraldehyde 3-phosphate dehydrogenase (GAPDH) was used as a loading control.…”

“…Importantly, our studies demonstrate that the polymerization of fibronectin fibrils can initiate cellular self-assembly under a range of adhesion conditions. Moreover, the attachment strength of initial/early adhesions may determine whether fibronectin fibrils and α5β1 integrins localize to cell-cell or cell-matrix adhesions, and function therein, possibly through changes in cell signaling that are associated with the transition to 3D [62]. …”

“…This method has been previously described in detail (10)(11)(12)13), and is also presented here as supplemental material. In brief, TST measures the intercellular binding energy of 3D spherical tissue aggregates under physiological conditions.…”

“…Some HDs were treated with vehicle, EMAPII (3 mg/ml), or 70-kD fragment of fibronectin (FN). After 48 hours in HDs, PB compaction was assessed as previously described (10)(11)(12)13). Briefly, phase-contrast images of compacted aggregates were captured, and each image was adjusted to optimize contrast.…”
