Thidiazuron suppresses breast cancer via targeting miR-132 and dysregulation of the PI3K–Akt signaling pathway mediated by the miR-202-5p–PTEN axis

Abstract: Chemo-resistance and metastatic disease development are the most common causes of breast cancer recurrence and death. Thidiazuron (TDZ) is a plant growth regulator, its biological role on human and animals has not been yet clarified. In the present study, we investigated the anticancer activity of this plant phytohormone on the drug resistant-triple negative breast cancer MDA-MB-231 cell line. Treatment of the breast cancer cells with TDZ (1-50 μM) caused more stressful environment and induced a significant in… Show more

“…Cell migratory properties affect tumorigenesis and metastasis. Various studies have targeted cancer cell migration and invasion as potent tools for controlling the progression of cancers [ 37 , 38 ]. In the current study, the cellular functional results revealed that EO (25 μg/mL) from A. lanatum exhibited a regulation of cell migration and invasion in HepG2 liver cancer cell lines ( Figure 3 and Figure 4 ).…”

“…The suppressive effects of BCL-2 and CYP-1A1 are interlinked in cancer prognosis through migratory action and tumorigenesis [ 45 ]. Angiogenesis involves regulatory functions played by BCL-2 and CASPASE-3 in reverse roles [ 38 , 46 ]. In this study, the suppression effect of EO on the cell migration process may have been mediated by the suppression of BCL-2 and NFκB markers ( Figure 5 ).…”

“…COX-2 is regulated by various epigenetic factors such as microRNAs (miRNAs), which play a major role in cancer management. These miRNAs are small non-coding nucleotides regulating posttranscription of mRNA in various biological processes including cancer [38]. COX-2 and its targeting miR-146a expression levels were considered as prognostic biomarkers for survival in various cancers including lung, breast, pancreatic, esophageal and gastric cancer [39][40][41][42].…”

“…The important downstream target that is affected by the miRNA/PTEN axis is PI3K/Akt signaling. For this purpose, thidiazuron activates the miRNA-202-5p/PTEN axis to suppress PI3K/Akt signaling, leading to breast cancer inhibition [302].…”
