Thiazolidinedione derivatives as novel agents against Propionibacterium acnes biofilms

Abstract: Aims: The aim of the present study was to determine the effect of two thiazolidinedione derivatives on Propionibacterium acnes biofilm formation in vitro and to assess their effect on the susceptibility of P. acnes biofilms towards antimicrobials. Methods and Results: The compounds were shown to have a moderate to strong antibiofilm activity when used in subinhibitory concentrations. These compounds do not affect P. acnes attachment but lead to increased dispersal of biofilm cells. This dispersal results in an… Show more

“…An anti-QS molecule from the class of thiazolidinediones, 5-octylidenethiazolidine-2,4-dione (TZD-8), has been found in our laboratory to be effective against several types of biofilm (Brackman et al, 2014;Srebnik et al, 2010), including those created by Candida (Feldman et al, 2014).…”

“…After 24 h biofilm formation, medium was removed, biofilms were washed with 100 μl PS and 100 μl of a staining solution (containing 3 μl of SYTO9 and 3 μl of propidium iodide in 1 ml of PS; Life technologies) were added. These plates were incubated in the dark for 15 min at room temperature and the biofilm was visualized with a Nikon C1 confocal laser scanning microscope (Nikon Benelux, Brussels, Belgium) as previously described50. Tests were performed on at least three wells for each condition and representative images are shown.…”

“…Prevention of the formation of biofilm is yet another avenue that has been proposed as a therapeutic modality. Brackman et al have used two thiazolidinedione drugs, (Z)-5-octylidenethiazolidine-2,4-dione (TZD8) and (Z)-5-decylidenethiazolidine-2,4-dione (TZD10), believed to play a role in inhibiting Autoindicer-2 dependent quorum sensing, the major mechanism by which P. acnes forms a biofilm [29].…”
