Thermoresponsive hydrogels from BSA esterified with low molecular weight PEG

Abstract: Bovine serum albumin (BSA) and low molecular weight polyethylene glycol (PEG) were reacted in a single-step reaction to synthesize translucent hydrogels with a sol-gel transition at temperatures between 37 and 40 C. Gelation occurred by aggregation of smaller assemblies of BSA-PEG precursors within minutes. The sol-gel transition concentration depended on the molecular weight of PEG only at temperatures below 35 C; above 45 C phase separation occurred and a precipitate formed. Microscopic examination showed th… Show more

“…As shown in Figure 1 B, BSA-based hydrogels were chosen as the model protein hydrogel due to their stable mechanical properties and excellent biocompatibility [ 33 , 34 , 35 , 36 , 37 ]. By mixing BSA protein solutions with N-hydroxysuccinimide-terminated 4-armed PEG (Mw: 20 kDa, named PEG), transparent hydrogels (denoted as BSA-PEG hydrogels) formed immediately in several minutes.…”

“…As described in a previous work, UV‐vis spectroscopy was used (Thermo Scientific Genesys 6 spectrometer; Thermo Scientific, Waltham, MA; quartz cuvettes with 10 mm path length) to measure the yield and concentration of the BSA moiety before the coating of the samples. The absorption coefficient for modified BSA used was 3,443,824 M −1 cm −1 (280 nm).…”

“…Albumin, which is a natural globular protein commonly found in blood plasma, is capable of showing a stimuli-responsive property in response to external stimuli like temperature and pH [24]. The thermo-responsive hydrogel made from bovine serum albumin (BSA) and low molecular weight polyethylene glycol (PEG) showed sol–gel transition near the body temperature and it was reported to be a promising drug delivery material [78]. The pH responsive hydrogel developed by free radical polymerization of methacrylate bovine serum albumin and N -isopropylacrylamide was used as a drug delivery system for caffeine and theophylline [79].…”
