Therapeutic potential of the Proprotein Convertase Subtilisin/Kexin family in vascular disease

Abstract: Proprotein convertase subtilisin/kexins (PCSKs) constitute a family of nine related proteases: PCSK1-7, MBTPS1, and PCSK9. Apart from PCSK9, little is known about PCSKs in cardiovascular disease. Here, we aimed to investigate the expression landscape and druggability potential of the entire PCSK family for CVD. We applied an integrative approach, combining genetic, transcriptomic and proteomic data from three vascular biobanks comprising carotid atherosclerosis, thoracic and abdominal aneurysms, with patient c… Show more

“…The following four gene expression profile datasets were retrieved from Gene Expression Omnibus (GEO) ( http://www.ncbi.nlm.nih.gov/geo ): GSE5180, GSE83675, GSE61128, and GSE2615 ( 11 13 ). In addition, we utilized self-test datasets, which included two aneurysm patients and two control groups.…”

“…A study categorized PCSK analysis into plaque expression, biochemical blood parameters, morphological characteristics, and patient symptoms. The cumulative data from each section suggested the feasibility of PCSK6 as a new therapeutic target, followed by PCSK5, PCSK7, and FURIN, all showing potential for CVD treatment within this family ( 143 ). This, of course, is also closely related to the treatment of AAA.…”

“…In patients with aortic dissections in a Korean population, for example, genomic alternations are found in a locus where the PCSK6 gene is located [ 126 ]. Genome-wide expression analysis indicates elevated PCSK6 expression in atherosclerotic plaques [ 127 , 128 ]. In cultured human monocytes and endothelial cells, PCSK6 expression and activity are increased by pro-atherogenic lipid oxidation products [ 129 ].…”
