Therapeutic Paint of Cidofovir/Sucralfate Gel Combination Topically Administered by Spraying for Treatment of orf virus Infections

Abstract: The aim of the research was to study a new cidofovir/sucralfate drug product to be used as a spray for treating the mucosal and/or skin lesions. The product, i.e., a water suspension of sucralfate (15% w/w) and cidofovir (1% w/w), combines the potent antiviral activity of the acyclic nucleoside phosphonate cidofovir ((S)-1-[3-hydroxy-2-(phosphonomethoxy)propyl]cytosine) and the wound healing properties of sucralfate gel (sucrose octasulphate basic aluminum salt). The product was characterized in vitro with res… Show more

“…Cidofovir has been tried as an anti-Orf virus for treatment of human Orf, with promising results, although current costs can be prohibitive for using it in lambs (De Clercq, 2002;Dal Pozzo et al, 2007). An experimental preparation containing sucralfate and cidofovir, in spray form, was shown to have efficacy in lambs, as it combined the antiviral activity of cidofovir and the wound healing properties of sucralfate (Sonvico et al, 2009).…”

“…Furthermore, cidofovir was also shown to reduce the amount of viable virus found in the lesions and thus could be critical to reducing the incidence of disease on farms that have a persistent problem with orf [159]. Recently, a spray formulation containing cidofovir/sucralfate gel was tested in vivo on lambs infected with ORFV; this gel proved to be active against the virus and was easy to use for the treatment of a large number of animals in the field [160].…”

“…Antinucleoside phosphonates, especially cidofovir, can be effective (De Clercq, 2002;Dal Pozzo et al, 2007), whilst use of a combination of cidofovir and sucralfate has also been proposed (Sonvico et al, 2009). …”
