SummaryAs anti-inflammatory treatments used in rheumatoid arthritis, such as glucocorticoids, often result in secondary detrimental effects on bone health, the objective of this study was to investigate the effects of oestrogen therapy (ET) on the development and activity of collagen-induced arthritis (CIA) in rats, with a focus on assessment of chondroprotective effects using biomarkers of type II collagen degradation. Forty female Lewis rats were allocated into four intervention groups: (i) control + vehicle; (ii) CIA + vehicle; (iii) CIA + ET; and (iv) CIA + prednisolone. During the 28-day intervention period we monitored body weight, time-point of disease onset, incidence of manifest disease and paw volume. Levels of the type II collagen degradation marker (CTX-II) were measured in serum. At euthanasia, hind paws were isolated, extracted for proteins and measured for the concentration of CTX-II. Matrix metalloproteinase (MMP) activity was evaluated using gelatinase zymography. Oestrogen treatment delayed the time-point of disease onset and reduced the incidence and degree of manifest immunoarthritis significantly, assessed by macroscopic evaluation of hind paw inflammation and paw volume. Measures of serum or tissue levels of CTX-II showed significantly reduced type II collagen degradation elicited by oestrogen treatment. In alignment, a decreased activity of MMP-2 and MMP-9 was found in the paw protein extracts. We have demonstrated that the anti-inflammatory effect of ET is linked to chondroprotective effects in an animal model of systemic immunoarthritis. As ET has positive rather than negative effects on bone health in contrast to prednisolone, these observations may be important for potential combination therapy.

Ginger and its extracts have been used traditionally as anti-inflammatory remedies, with a particular focus on the medicinal properties of its phenolic secondary metabolites, the gingerols. Consistent with these uses, potent anti-arthritic effects of gingerol-containing extracts were previously demonstrated by our laboratory using an experimental model of rheumatoid arthritis, streptococcal cell wall (SCW)-induced arthritis. In this study, anti-inflammatory effects of ginger's other secondary metabolites, the essential oils (GEO), which contain terpenes with reported phytoestrogenic activity, were assessed in female Lewis rats with SCW-induced arthritis. GEO (28 mg/kg/d ip) prevented chronic joint inflammation, but altered neither the initial acute phase of joint swelling nor granuloma formation at sites of SCW deposition in liver. Pharmacologic doses of 17-β estradiol (200 or 600 μg/kg/d sc) elicited the same pattern of anti-inflammatory activity, suggesting that GEO could be acting as a phytoestrogen. However, contrary to this hypothesis, GEO had no in vivo effect on classic estrogen target organs, such as uterus or bone. En toto, these results suggest that ginger's anti-inflammatory properties are not limited to the frequently studied phenolics, but may be attributable to the combined effects of both secondary metabolites, the pungent-tasting gingerols and as well as its aromatic essential oils.

The aim of this study was to investigate the development of collagen-induced arthritis (CIA) in male and female Wistar and Lewis rats and establish relationships between clinical status and the levels of serum oxidative products. CIA was induced in 64 rats divided randomly into three groups. Animals of the 1st group received one 0.1 ml injection of bovine type II collagen (CII, in dose of 0.1 mg/rat) emulsified in incomplete Freund's adjuvant (IFA). Rats of the 2nd group were immunized twice: on day 0 (dose of CII 0.1 mg/rat) and day 7 (dose of CII 0.05 mg/rat), and the animals of the 3rd group received one 0.1 ml injection of emulsion consisting of CII (0.1 mg/rat) in IFA and muramyldipeptide (MDP, in dose of 3 mg/ml). Serum oxidative products such as malondialdehyde (MDA), anti-oxidative enzyme catalase (CAT), total antioxidant activity (AOA), and arthritic profiles based on paw swelling, development of polyarthritis and histological changes in joints were measured in rats with CIA. Our findings have demonstrated that CIA develops in both sexes of Wistar and Lewis rats and the disease that occurs is an inflammatory erosive arthritis. Examination of the clinical course of the disease and the subsequent histological analysis disclosed a slight less aggressive disease in Wistar than in Lewis rats and in female than in male animals. The most severe arthritis developed in the 3rd group of male Lewis rats. CIA induced significant changes in the parameters of the pro-/antioxidant status of serum. Comparison of the oxidative statuses of both strains of rats with CIA and those of healthy animals revealed more elevated MDA levels in the serum of Wistar than Lewis rats. More evidently in the serum of these rats the level of total AOA was reduced, especially in the 3rd group of animals. More susceptible to CIA, Lewis rats showed a lower MDA production as compared with Wistar animals, and the lowest CAT activity in the serum of these rats was observed. In conclusion, attenuated inflammatory response and pathomorphological changes in joints were more observed in female animals of both strains. Male Lewis rats were most susceptible to CIA. On the basis of increased lipid peroxidation and decreased levels of AOA and enzyme CAT activity, CIA rats are subject to oxidative stress.
