Therapeutic effect of prostaglandin E1 in monocrotaline-induced pulmonary arterial hypertension rats

Abstract: Pulmonary arterial hypertension (PAH) is a severe pulmonary vascular disease characterized by sustained increase in pulmonary arterial pressure and excessive thickening and remodeling of distal small pulmonary arteries. During disease progression, PAH include increase in mean pulmonary arterial pressure, right ventricular (RV) enlargement, increased pulmonary vascular resistance, and smooth muscle hypertrophy in pulmonary arterioles. Several anti-PAH therapies targeting various pathways involved in PAH progres… Show more

“…Because PGE1 is a well-known potent vasodilator (Hasegawa and Ichioka, 2015; Lee, 2017). To investigate whether the enhancement of CBF and cognitive functions by PGE1 was attributed to vascular alteration, we examined the effect of PGE1 on vertebral arteries (VAs).…”

“…Activation of the EP3 receptor (so-called “inhibitory”) reduces the level of cyclic adenosine monophosphate (cAMP), whereas the EP2 and EP4 receptors (so-called “relaxing”) increase the level of cAMP [ 95 , 96 ]. PGE1 has strong vasodilatory, anti-inflammatory, anti-proliferative properties, is a strong platelet aggregation inhibitor, stimulates smooth muscles, reduces platelet hyperactivity and thromboxane A2 (TXA2) level, reduces free Ca 2+ levels in vascular smooth myocytes [ 97 , 98 ]. It has been found that PGE1 influences the maintenance of renal hemodynamic homeostasis by regulating blood flow and distribution as well as electrolyte and water excretion [ 99 ].…”
