Background: Acute pancreatitis (AP) is a common inflammatory disorder of digestive system. Mesenchymal stem cells (MSCs) and Wheat germ oil (WGO) could improve AP through their anti-inflammatory and antioxidant effects. Objective: Evaluate and compare the possible therapeutic effects of Bone marrow derived Mesenchymal Stem Cells (BMSCs) versus WGO on AP. Materials and Methods: 47 adult male albino rats were divided into 4 groups. Control group I (no.=12). AP was induced in the remaining 35 rats by a single intra peritoneal (I.P) injection of L-arginine (250 mg/100g). 5 rats died within the 1st hour after AP induction, the rest were divided randomly into group II (AP group; no.=10) that received no treatment, group III (BMSCs group; no.=10) and group IV (WGO group; no.=10). One hour after AP induction, group III was injected I.P. by 1ml of PKH26 labeled BMSCs (1x106 cells/ml) and group IV received WGO in a dose of 3 ml/kg body weight by oral gavage every 24 h for 3 successive days. Blood samples were collected 24 hours and on the 4 th day after AP induction for biochemical assessment of serum amylase, lipase, interleukin-1β and interleukin-10. Then, animals were sacrificed and specimens from the pancreas were prepared for Hematoxylin and eosin (HandE) stain and immune-histochemical staining using inducible Nitric Oxide Synthase (iNOS) and insulin antibodies. Morphometric measurements using image analyzer were done. Results: Group II showed extensive pancreatic damage associated with increase in serum amylase, lipase and interleukin-1B levels and reduction in interleukin-10 level. A significant increase in the area % of iNOS immunostaining and non-significant change in insulin immunostaining were detected. On the other hand, BMSCs group and WGO group showed improvement in the biochemical, histological and immunohistochemical results with better results in BMSCs group. Conclusion: BMSCs possess better therapeutic efficacy in treating AP compared with WGO.

Introduction: Hyperlipidaemia is a dangerous condition that may lead to atherosclerosis and various cardiovascular diseases. Different drugs are taken to reduce hyperlipidaemia such as Statins. Recently various side effects for statins have been detected clinically that actually restrict their prescription. Resveratrol is a natural phenol and is produced naturally by several plants and proved to have both antioxidant & anti-inflammatory actions. Aim of the Study: This research has been aimed to investigate the effect of Atorvastatin on the pancreas and the probable protective action of Resveratrol. Materials and Methods: Forty adult male albino rats were divided into four equal groups and were received drugs orally as a single daily dose for one week. Control group (I): received 1ml normal saline. Resveratrol group (II): received dose 30 mg/ kg. Atorvastatin group (III): received dose 80 mg/Kg. Group IV: received Resveratrol concomitant with Atorvastatin with the same doses as pervious groups.All rats were sacrificed one day following the last dose and blood samples were gathered for biochemical study of serum amylase and lipase. Pancreas specimens were obtained and processed for histological and immunohistochemical study. Results: Atorvastatin-treated rats showed degeneration of pancreatic acini. The acinar cells showed cytoplasmic vacuoles with dilated RER and depleted zymogen granules. Also, inflammatory cell infiltration, dilated congested blood vessels with hemorrhage and marked deposition of collagen fibers were observed. The islets of Langerhans showed apparent reduced cells number. α cells and to more extent β cells revealed degeneration. The immunohistochemical results revealed marked increase in vascular endothelial growth factor (VEGF) and caspase-3 expressions. The biochemical study revealed remarkable elevation in the serum amylase and lipase. The concomitant administration of Resveratrol with Atorvastatin minimizes these changes. Conclusion:Atorvastatin has been proved to induce pancreatic histological changes and these changes can be attenuated by Resveratrol when given concomitantly with it.

Inflammation is a normal part of the immune response to injury or infection but its dysregulation promotes the development of inflammatory diseases, which cause considerable human suffering. Nonsteroidal anti-inflammatory agents are the most commonly prescribed agents for the treatment of inflammatory diseases, but they are accompanied by a broad range of side effects, including gastrointestinal and cardiovascular events. The renin-angiotensin system (RAS) is traditionally known for its role in blood pressure regulation. However, there is increasing evidence that RAS signaling is also involved in the inflammatory response associated with several disease states. Angiotensin II increases blood pressure by binding to angiotensin type 1 (AT ) receptor, and direct renin inhibitors, angiotensin-converting enzyme (ACE) inhibitors and AT receptor blockers (ARBs) are clinically used as antihypertensive agents. Recent data suggest that these drugs also have anti-inflammatory effects. Therefore, this review summarizes these recent findings for the efficacy of two of the most widely used antihypertensive drug classes, ACE inhibitors and ARBs, to reduce or treat inflammatory diseases such as atherosclerosis, arthritis, steatohepatitis, colitis, pancreatitis, and nephritis.
