Antiproliferative Activity of Artemisia asiatica Extract and Its Constituents on Human Tumor Cell Lines

The extract of Artemisia asiatica herb with antiproliferative activity against four human tumor cell lines (A2780, A431, HeLa, and MCF7) was analyzed by the MTT assay, and bioassay-directed fractionation was carried out in order to identify the compounds responsible for the cytotoxic activity. Guaianolide (1-4), seco-guianolide (5), germacranolide (6) and eudesmanolide sesquiterpenes (7), monoterpenes (8, 9), including the new compound artemisia alcohol glucoside (8), and flavonoids (10-16) were isolated as a result of a multistep chromatographic procedure (CC, CPC, PLC, and gel filtration). The compounds were identified by means of UV, MS, and NMR spectroscopy, including (1)H-and (13)C-NMR, (1)H-(1)H COSY, NOESY, HSQC, and HMBC experiments. The isolated compounds 1-16 were evaluated for their tumor cell growth-inhibitory activities on a panel of four adherent cancer cell lines, and different types of secondary metabolites were found to be responsible for the cytotoxic effects of the extract. Especially cirsilineol (13), 3β-chloro-4α,10α-dihydroxy-1α,2α-epoxy-5α,7αH-guai-11(13)-en-12,6α-olide (3), and iso-seco-tanapartholide 3-O-methyl ester (5) exerted marked cytotoxic effects against the investigated cell lines, while jaceosidin (12), 6-methoxytricin (15), artecanin (2), and 5,7,4',5'-tetrahydroxy-6,3'-dimethoxyflavone (14) were moderately active. All the sesquiterpenes and monoterpenes are reported here for the first time from this species, and in the case of artecanin (2), 3α-chloro-4β,10α-dihydroxy-1β,2β-epoxy-5α,7αH-guai-11(13)-en-12,6α-olide (4), ridentin (6), and ridentin B (7), previously unreported NMR spectroscopic data were determined.

Background/Aims: Acute gastric injury by alcohol or indomethacin has been reported to be prevented by DA-9601, an extract of the herb Artemisia asiatica. Ghrelin, an endogenously produced gastrointestinal peptide hormone, has also been demonstrated to play a role in gastric mucosal defense. The aim of this study was to investigate the effects of DA-9601 on ghrelin in an acute gastric injury model induced by alcohol or indomethacin. Methods: A total of 140 SpragueDawley rats were divided into two groups, a placebo group and a DA-9601-pretreated group. Thirty minutes later, half of the rats in each group received ethanol injury and the other half received indomethacin injury. Levels of serum ghrelin and gastric mucosal ghrelin mRNA were measured by ELISA and RT-PCR, respectively. Results: Immediately after ethanol administration, ghrelin increased in both groups pretreated with DA-9601 and placebo. However, the increase occurred more rapidly and was higher in the DA-9601-pretreated rats than in the controls that did not receive DA-9601-pretreatment. Similarly, from 30 minutes to 2 hours after indomethacin administration, the DA-9601-pretreated rats showed a significant increase in serum and gastric mucosal ghrelin concentrations, whereas placebo-pretreated rats showed only a mild increase. Conclusions: DA-9601 potentiates the endogenous production and secretion of ghrelin in acute gastric injury models induced by ethanol or indomethacin. (Gut Liver 2011;5:52-56) Key Words: DA-9601; Ghrelin; Acute gastric injury INTRODUCTIONThe gastric mucosa defends its structural integrity against many noxious substances such as alcohol and nonsteroidal anti-inflammatory drugs (NSAIDs) via pre-epithelial, epithelial and sub-epithelial factors that include the mucus barrier, bicarbonate secretion, epithelial tight junctions, prostaglandin (PG) synthesis, cell restitution, and mucosal blood flow.1 Alcohol ingestion may lead to gastric mucosal lesions by penetrating the gastric mucosa and causing mucosal inflammation, erosions and ulcer formation. 2-4 Alcohol-induced gastric injury is mediated by free radicals, which is supported by the fact that ethanol administration depletes cells of major antioxidants such as superoxide dismutase, catalase, and glutathione peroxidase. Gastric injury induced by NSAIDs such as indomethacin is mediated by nonselective inhibition of the cyclooxygenase pathways, which results in decreased PG synthesis. DA-9601, an extract of Artemisia asiatica, has been reported to have anti-inflammatory, antioxidative and cytoprotective effects and therefore to play a protective role in experimentallyinduced gastrointestinal damage and hepatic and pancreatic lesions. “…Although we presented the detailed data about DA-9601 treatment, ethanol extracts of Artemisia asiatica [ 11 ], among several gastroprotectants [ 8 – 10 ], which is one of gastroprotectants prescribed popularly for the treatment of gastritis and peptic ulcer disease in Korea, possessing actions like anti-inflammatory, cytoprotective, restorative, and antioxidative actions [ 11 , 21 – 24 ], we repeated the same experiment with other gastroprotectants available in clinic like rebamipide and ecabet sodium, and found that the overall rates of resistance to ulcer recurrence experiment according to gastric cytoprotectants were 2/12 (16.7%) with DA-9601, 3/12 (25.0%) with rebamipide, and 3/11 (27.2%) with ecabet sodium, respectively. Summary Quality of ulcer healing (QOUH) is defined as ideal ulcer healing featuring with the fine granular ulcer scar, high functional restoration and the resistance to recurrence. This study was designed to compare the rates of QOUH achievement in rat gastric ulcer model between acid suppressant treated group and gastroprotectant treated group accompanied with elucidations of molecular mechanisms. Serosal injection of acetic acids for generating gastric ulcer and intraperitoneal (ip) injection of recombinant interleukin 1-beta (IL-1β) for recurring healed ulcer was done in SD rats. The 72 rats were divided into three groups according to treatment as follows; Group I, no further treatment, Group II, 8 weeks treatment of omeprazole, and Group III, 8 weeks of gastroprotectant treatment. IL-1β was administered for ulcer recurrence after 28 weeks of acetic acid injection. At four weeks after gastric ulcerogenesis, 58.3% (7/12) of active gastric ulcer were converted to healing stage in Group III, but 16.7% (2/12) in Group II and none in Group I, for which significant levels of epidermal growth factor, mucin, and pS2/trefoil peptide1 were contributive to these accelerated healings of Group III. ip injections of rIL-1β (200 µg/kg) at 28 weeks after acetic acid injection led to 100% of ulcer recurrence in Group I and 75.0% in Group II, but only 16.7% of Group III rats showed ulcer recurrence. Significantly attenuated levels of inflammatory cytokines including IL-2, transforming growth factor-alpha (TNF-α), cyclooxygenase-2 (COX-2), nitrotyrosine were responsible for the resistance to ulcer recurrence in Group III. Conclusively, gastroprotectant might be prerequisite in order to achieve ideal QOUH through significant inductions of remodeling.
