TheHIS4 gene from the yeastKluyveromyces lactis

Abstract: The Kluyveromyces lactis HIS4 gene was cloned by complementation of a Saccharomyces cerevisiae his4 mutant. Sequence analysis revealed a 2388 bp open reading frame encoding a single polypeptide predicted to encompass three distinct enzymatic activities (phosphoribosyl‐AMP cyclohydrolase, phosphoribosyl‐ATP pyrophosphohydrolase and histidinol dehydrogenase). This structural organization is strikingly similar to that of the His4 proteins from S. cerevisiae and Pichia pastoris. Transcript analysis detected a sing… Show more

“…TFIIFα has important roles during transcription initiation, elongation and even pausing (Gong et al, 1993;Landick, 2006;Price et al, 1989;Rossignol et al, 1999). Based on its role in different steps of transcription, TFIIFα, either alone or associated with other TFs, might be influencing transcription start site selection (Freire-Picos et al, 2005;Khaperskyy et al, 2008) or preventing RNA PolII (which was not certified by peer review) is the author/funder. “…The 1-kb fragment obtained by PCR, with OMLPR6 5 0 -TAGGATCCACTG CAAATCAAGC-3 0 and OML15 5 0 -CATGCCCATAAAG GG-3 0 and pM756 [7] as template, was used as the KlHIS4 probe. The KlLEU2 probe was also obtained by PCR, with OMLKL1 5 0 -CCTTAATAGCTTCGTCAG-3 0 and OMLKL2 5 0 -TTTAACGCATCCTACATTTG-3 0 using NRRL-Y1140 genomic DNA as template.…”

“…After cloning the KlHIS4 gene by complementation, it was found to encode a protein with a 69% identity and 81% similarity to the multi‐enzymatic protein encoded by the S. cerevisiae HIS4 gene [18]. Herein we report the sequence of the promoter region which is responsible for its transcriptional regulation and necessary in order to allow histidine auxotrophy complementation in S. cerevisiae .…”
