The α7β1 integrin in muscle development and disease

Abstract: The alpha7beta1 integrin is a laminin receptor on the surface of skeletal myoblasts and myofibers. Alternative forms of both the alpha7 and beta1 chains are expressed in a developmentally regulated fashion during myogenesis. These different alpha7beta1 isoforms localize at specific sites on myofibers and appear to have distinct functions in skeletal muscle. These functions include the migration and proliferation of developing myoblasts, the formation and integrity of neuromuscular and myotendinous junctions, a… Show more

“…The ␣ 7 ␤ 1 -integrin is the predominant laminin-binding integrin in cardiac and skeletal muscle (7). Mutations in the ␣ 7 -integrin gene cause congenital myopathy in both humans and mice (8)(9)(10).…”

“…In mouse, ADAM12 positively modulates expression of integrin ␣ 7 (Cao et al, 2003;Moghadaszadeh et al, 2003), which has functions in skeletal muscle development and disease (Burkin and Kaufman, 1999). ␣ 7 ␤ 1 integrin is mainly involved in mpc adhesion and migration on the matrix component laminin (Yao et al, 1996;Crawley et al, 1997;Blanco-Bose and Blau, 2001) and in myotendinous junction formation (Nawrotzki et al, 2003).…”

“…Therefore, we have established a cell-free screening assay in which we used a recombinantly expressed soluble ␣ 7 ␤ 1 integrin heterodimer and its binding to laminin-1. Within the muscle, ␣ 7 ␤ 1 is highly concentrated at the myotendinous and neuromuscular junctions, which are essential for the transduction of mechanical forces and nerve signals, respectively (12). Because ␣ 7 X2␤ 1 integrin seems to be the only isoform involved in laminininduced acetylcholine receptor recruitment into neuromuscular junctions (12) and is the predominant isoform in adult skeletal muscle, the splice variant X2 was chosen for recombinant production.…”

A high yield expression of soluble ␣ 7 X2␤ 1 integrin was achieved by coexpression of the murine ␣ 7 X2 integrin ectodomain with the human ␤ 1 integrin ectodomain in Drosophila Schneider's cells.…”
