The Zinc Finger Transcription Factor Sp9 Is Required for the Development of Striatopallidal Projection Neurons

Abstract: SUMMARY Striatal medium-sized spiny neurons (MSNs), composed of striatonigral and striatopallidal neurons, are derived from the lateral ganglionic eminence (LGE). We find that the transcription factor Sp9 is expressed in LGE progenitors that generate nearly all striatal MSNs, and that Sp9 expression is maintained in postmitotic striatopallidal MSNs. Sp9 null mice lose most striatopallidal MSNs due to decreased proliferation of striatopallidal MSN progenitors and increased Bax-dependent apoptosis, whilethe deve… Show more

“…Sp9 was reported to be widely expressed in the ganglionic eminences (GEs) and in all striatopallidal MSNs. The expression pattern observed in this study is consistent with that of (Zhang et al, ).…”

“…SP8 is expressed in the SVZ of the dLGE and dCGE (Ma et al, ) and SP9 is widely expressed in the SVZ of the GE (Guo et al, ; Liu et al, ; Long et al, ; Xu et al, ; Zhang et al, ). In this study, we found SP8/9 were highly co‐expressed in the dCGE SVZ.…”

“…Studies in recent years have begun to shed light on the molecular genetic mechanisms that control the development of the direct and indirect SPN pathways. In this respect, Ikaros and Sp9 are required for the normal differentiation and survival of the indirect SPNs (Martin-Ibanez et al, 2010; Zhang et al, 2016). Conversely, Ebf1 (Garel et al, 1999; Lobo et al, 2006, 2008) and the LIM homeodomain protein, Isl1 (Ehrman et al, 2013; Lu et al, 2014) have been implicated in the development of the direct pathway (i.e.…”

“…Moreover, Ctip2 and Foxp1 have been shown to be required for the normal differentiation of SPNs (Arlotta et al, ; Bacon et al, ). The zinc finger transcription factor Sp9 is expressed by many subventricular zone cells of the LGE, however, it's expression resolves to the iSPNs at postnatal stages and is required for the proliferation and postnatal survival of this SPN subtype (Zhang et al, ). Conversely, the early B cell factor Ebf1 , is known to be required for the normal development and connectivity of dSPNs, particularly in the matrix compartment (Garel, Marin, Grosschedl, & Charnay, ; Lobo, Karsten, Gray, Geschwind, & Yang, ; Lobo, Yeh, & Yang, ).…”
