The yeast aquaglyceroporin Fps1p is a bidirectional arsenite channel

Abstract: a b s t r a c tThe stress-activated kinase Hog1p mediates arsenic tolerance by decreasing arsenite influx through the aquaglyceroporin Fps1p in Saccharomyces cerevisiae. Unexpectedly, we found that overexpression of FPS1 increased arsenite tolerance suggesting a physiological role of Fps1p in arsenic detoxification. Consistently, during arsenite treatment transcription of FPS1 gene was strongly upregulated, while Fps1p was not degraded and remained localized to the plasma membrane. Moreover, deletion of FPS1 g… Show more

“…For copper uptake, the liquid SC-U/Glu yeast culture (OD600 ϳ 0.5) was supplied with 100 M CuCl 2 and subsampled after 1, 2, 4, and 8 h of growth in the presence of metal excess. The intracellular copper content was determined essentially as described earlier (24,25) using the Atomic Absorption Spectrophotometer 3300 (PerkinElmer Life Sciences).…”

“…Aquaporins mediate non specific uptake of arsenite and mutants in the corresponding genes are usually arsenite resistant [7], [8], [9], [10], [61]. The only gene coding for this protein in Synechocystis ( aqpZ; slr2057 ) was repressed after both arsenate and arsenite (10 fold in arsenate and 6 fold in arsenite; Table S1).…”

“…The A. niger genomic sequences were obtained from the Central Aspergillus Data Repository (CADRE) (www.cadre-genomes.org.uk/Aspergillus_niger/). The putative arsenic homeostasis genes in Aspergillus niger were found by comparing gene and protein sequences with those of S. cerevisiae (12,35,39,49,61,62,66) using nucleotide-BLAST and protein-BLAST from the National Center for Biotechnology Information (http://blast.ncbi.nlm.nih .gov/Blast.cgi).…”
