The Ycf48 accessory factor occupies the site of the oxygen-evolving manganese cluster during photosystem II biogenesis

Abstract: Robust oxygenic photosynthesis requires a suite of accessory factors to ensure efficient assembly and repair of the oxygen-evolving photosystem two (PSII) complex. The highly conserved Ycf48 assembly factor binds to the newly synthesized D1 reaction center polypeptide and promotes the initial steps of PSII assembly, but its binding site is unclear. Here we have used cryo-electron microscopy to determine the structure of a cyanobacterial PSII D1/D2 reaction center assembly complex with Ycf48 attached. Ycf48, a … Show more

“…We probed our fractions for six proteins involved in PSII assembly (Figure 2). HCF136, which acts very early in the assembly of D1 into the PSII reaction center (Meurer et al, 1998;Plücken et al, 2002;Zhao et al, 2023), was distributed among the refined margin, curvature, and stromal lamellae fractions, consistent with the prior localization of HCF136 to crude margins and stromal lamellae (Garcia-Cerdan et al, 2019). HCF244, OHP2, and RBD1 also mediate early steps in assembly of the PSII reaction center and had previously been localized to crude margin fractions (Knoppova et al, 2014;Hey and Grimm, 2018;Garcia-Cerdan et al, 2019;Li et al, 2019;Maeda et al, 2022;Calderon et al, 2023).…”

“…We therefore propose that LHL4 binding to the PSII monomer during exposure to HL promotes the formation of an intermediate shielded form that protects the complex from photoinhibition. The structural mechanism is reminiscent of that recently proposed for the PSII oxygen-evolving complex, where the Ycf48 assembly factor binds to the PSII monomer, promoting the initial steps of PSII assembly in cyanobacteria (49).…”

“…In parallel, the newly synthesized D1 precursor interacts with PsbI that has already been produced. PsbI-D1 (D1 mod ) is then combined with the D2-Cyt b 559 complex to form the reaction center (RCII) (Dobakova et al, 2007; Zhao et al, 2023). This is followed by proteolytic processing of the D1 precursor at its C-terminus (Anbudurai et al, 1994).…”
