The WNT target SP5 negatively regulates WNT transcriptional programs in human pluripotent stem cells

Abstract: The WNT/β-catenin signaling pathway is a prominent player in many developmental processes, including gastrulation, anterior–posterior axis specification, organ and tissue development, and homeostasis. Here, we use human pluripotent stem cells (hPSCs) to study the dynamics of the transcriptional response to exogenous activation of the WNT pathway. We describe a mechanism involving the WNT target gene SP5 that leads to termination of the transcriptional program initiated by WNT signaling. Integration of gene exp… Show more

“…The identification of motif consensus among the Sp5-enriched regions revealed the same core consensus motif GG(A/T/C)GG, centrally enriched within both the HySp5 and ZfSp5a bound sequences, very similar to the motif described for the human Sp5 protein (Huggins et al, 2017). Two other consensus motifs were found in the HySp5 dataset but neither in the ZfSp5a dataset, nor in the human or mouse Sp5 ChIPseq analyses (Figure 7D -Figure Supplement 9).…”

“…Human embryonic stem (hES) cell H9 (WA09, NIH Registration number 062) cells were obtained from WiCell. Cells were maintained in Essential 8 (E8) media (Chen et al, 2011), with minor modifications, as previously described (Huggins et al, 2017). Plasmids encoding pools of shRNAs for FZD9 and WNT9A were obtains from ABM.…”

“…construct. APEX2-mediated proximity labeling was carried out essentially as described (Hung et al, 2016;Lam et al, 2015). Briefly, confluent 150mm plates of CMV:Fzd9b-5GS-APEX2 cells were treated with biotin-phenol for a total of 30 minutes each, ending at the time of hydrogen peroxide treatment.…”
