The Wnt Antagonist Dickkopf-1 Promotes Pathological Type 2 Cell-Mediated Inflammation

Abstract: Summary Exposure to a plethora of environmental challenges commonly triggers pathological type 2 cell-mediated inflammation. Here we report the pathological role of the Wnt antagonist Dickkopf-1 (Dkk-1) upon allergen challenge or non-healing parasitic infection. The increased circulating amounts of Dkk-1 polarized T cells to T helper 2 (Th2) cells, stimulating a marked simultaneous induction of the transcription factors c-Maf and Gata-3, mediated by the kinases p38 MAPK and SGK-1, resulting in Th2 cell cytokin… Show more

“…Previous studies demonstrated thrombocytes (or platelets) or cancer cells as sources of DKK‐1 upon exposure to external stimuli (e.g. parasites or allergens) or alterations in cancerous cells . Notably, membrane‐bound TGF‐ β is also highly expressed in thrombocytes and Treg cells similar to DKK‐1 .…”

“…We observed that the gene expression of IL‐2‐mediated signalling inhibitory protein Suppressor of Cytokine Signalling‐1 was increased by DKK‐1 (data not shown). DKK‐1 may inhibit Th1 cell differentiation since our previous study showed that DKK‐1 antagonizes the expression of interferon‐ γ expression by increasing Gata‐3 …”

“…Given that Wnt5a supports a proinflammatory cytokine signature (46), its potential for protection against parasite infection may also involve prevention of the predominantly Th2 signature that sustains infection (1). Recently, high serum levels of the Wnt antagonist Dkk1 have been correlated with a predominantly Th2 phenotype during the onset of experimental cutaneous leishmaniasis (47). Although cutaneous leishmaniasis and visceral leishmaniasis follow different disease courses, the overall negative correlation between Wnt signaling and infection supports the relevance of Wnt signaling in host defense against parasite infections.…”
