The WD40 Protein BamB Mediates Coupling of BAM Complexes into Assembly Precincts in the Bacterial Outer Membrane

Abstract: The β-barrel assembly machinery (BAM) complex is essential for localization of surface proteins on bacterial cells, but the mechanism by which it functions is unclear. We developed a direct stochastic optical reconstruction microscopy (dSTORM) methodology to view the BAM complex in situ. Single-cell analysis showed that discrete membrane precincts housing several BAM complexes are distributed across the E. coli surface, with a nearest neighbor distance of ∼200 nm. The auxiliary lipoprotein subunit BamB was cru… Show more

“…Consistent with this, dolP shows synthetic lethality with the gene encoding the periplasmic chaperone SurA, leading to suggestions of a role for DolP in OMP biogenesis [29][30][31] . However, we were unable to demonstrate a direct interaction between DolP and the BAM complex, and no such interaction has been seen in the extensive studies evaluating the subunit composition and multimeric states of the BAM complex [32][33][34][35] or in similar studies in N.…”

“…Evidence from in vivo work suggests that SurA may be involved in direct delivery of OMP substrates to BAM [19][20][21][22][23], although the molecular mechanism by which this occurs remains unresolved.…”

“…Although OMPs are a major component of the OM, there is scarce information about their localization and mobility (18). In E. coli, naturally abundant or overexpressed OMPs show an even distribution (19,20), whereas less abundant proteins have been shown to localize in patches (18,19,21,22) or in particular cases at the cell pole (23). These patches have been suggested to be organized in spirals (24).…”

“…Integration of newly synthesized OMPs in the OM is accomplished by the BAM complex (27). BAM complexes have been suggested to be arranged in groups of 4 to 6 distributed across the cell, and at least one-third of the OMP patches have colocalized with a BAM complex (21,22). This organization has led to the suggestion that the BAM complexes form functional assembly precincts in the OM.…”
