The Wax Ester Synthase/Acyl Coenzyme A:Diacylglycerol Acyltransferase from Acinetobacter sp. Strain ADP1: Characterization of a Novel Type of Acyltransferase

Abstract: The wax ester synthase/acyl coenzyme A (acyl-CoA):diacylglycerol acyltransferase (WS/DGAT) catalyzes the final steps in triacylglycerol (TAG) and wax ester (WE) biosynthesis in the gram-negative bacterium Acinetobacter sp. strain ADP1. It constitutes a novel class of acyltransferases which is fundamentally different from acyltransferases involved in TAG and WE synthesis in eukaryotes. The enzyme was purified by a three-step purification protocol to apparent homogeneity from the soluble fraction of recombinant … Show more

“…in the dogfish Squalus acanthias) (62). Triglycerides in general are well known storage compounds of carbon and energy, which are used by several eukaryotes like mammals and plants (12), but have only rarely been found in prokaryotes, which in some cases seem to use polyhydroxyalkanoates instead (63)(64)(65)(66)(67)(68).…”

“…The third type is the Acinetobacter calcoaceticus WS enzyme, which shows both WS activity and DGAT activity. This bi-functional protein is unrelated to the mammalian or jojoba WS enzymes, and it shows preference for C14 and C16 acyl-CoA, together with C14 to C18 alcohols (Kalscheuer and Steinbuchel, 2003;Stoveken et al, 2005). Finally Li et al (2008) described the WS/DGAT enzyme that catalyzes the biosynthesis of wax esters in Arabidopsis.…”

“…Ce projet nommé « Diet Coli » (postiche du « Diet Cola ») est réalisable par expression de l'enzyme DGAT (diacylglycerol acyltransferase). L'équipe a montré que l'expression de la DGAT de Acinetobacter ADP1 [5] par E. coli induit en effet la synthèse de triglycérides, dans des proportions encore augmentées si des acides gras sont présents dans le milieu. La conception de la bactérie multicellulaire synthétique a fait l'objet d'une étude théorique et expérimentale poussée.…”
