The VHL/HIF axis in clear cell renal carcinoma

Abstract: Inactivation of the VHL tumor suppressor protein (pVHL) is a common event in clear cell renal carcinoma, which is the most common form of kidney cancer. pVHL performs many functions, including serving as the substrate recognition module of an ubiquitin ligase complex that targets the alpha subunits of the heterodimeric HIF transcription factor for proteasomal degradation. Deregulation of HIF2α appears to be a driving force in pVHL-defective clear cell renal carcinomas. In contrast, genetic and functional studi… Show more

“…Finally, loss of VHL results in the accumulation of HIF-α subunits, leading to activation of the HIF-2 (and possibly HIF-1) transcription factors (25). To determine whether Vhl loss may similarly induce Hif in the mouse, we performed qRT-PCR assays for a panel of Hif-2 and Hif-1 target genes, including VEGF, PAI-1, Glut1, IGFBP3, and PGK1 (26).…”

“…The role of pVHL has been extensively studied [5]. pVHL is a multifunctional protein whose function has been clearly linked to kidney carcinogenesis due to its role in the polyubiquitination process.…”
