The vasodilatory effect of ketamine is independent of the N-methyl-D-aspartate receptor: lack of functional N-methyl-D-aspartate receptors in rat mesenteric artery smooth muscle

Abstract: These results suggest that the NMDAr is not functional in vascular smooth muscle, and the vasodilatory action of ketamine is independent of the NMDAr in the rat mesenteric artery.

“…This is in line with EEG studies that have demonstrated increases in highfrequency and decreases in low-frequency neural oscillations in humans (80) and in mice (81). In addition, resting-state positron emission tomography studies have demonstrated that ketamine-induced increase in regional blood flow is counterintuitively associated with reduced oxygen extraction (82,83), possibly related to the direct vascular effect of ketamine (84).…”

“…The Institutional Animal Care and Use Committee of Konkuk University approved this study. Mesenteric arterial rings were prepared as previously described [16]. Male Sprague-Dawley (SD) rats (10- to 11-week-old) were exsanguinated by cutting the carotid arteries under deep ketamine-xylazine anesthesia or after exposure to rising concentrations of carbon dioxide.…”

“…The isometric tension of arterial rings was measured as previously described [16]. The arterial rings were mounted vertically on two L-shaped stainless steel wires in a 3 mL tissue chamber.…”

“…La respuesta hiper tensiva no parece estar relacionada a estos receptores NMDA, ni centrales ni periféricos. En estos últimos, experi mentalmente en arterias mesentéricas in vitro de rata, la ketamina actúa pro duciendo más bien vasodilatación (29) , mecanismo no esperado en la respuesta hipertensiva y que es independiente de los receptores NMDA que no parecen ser funcionales en la estructura muscu lar lisa mesentérica.…”
