The use of synchrotron infrared microspectroscopy to demonstrate the effect of intense pulsed light on dermal fibroblasts

Abstract: Intense pulsed light (IPL) has been used extensively to treat striae distensae (SD). Twenty-four patients (21 females and 3 males) with age ranging between 15-42 years with a mean of 24.2 ± 5.89 years were included in this study. All patients were treated with IPL for five sessions and were given 4-week interval between the sessions. Formalin-fixed, paraffin- embedded skin biopsies were cut into 5-μm-thick sections from patients before and after treatments. They were stained with hematoxylin and eosin (H&E) an… Show more

“…S v , indicating depth of atrophic lesions, decreased from baseline mean score of À285.0 AE 63.66 lm to À194.6 AE 37.32 lm at 1-month FU, with À203.8 AE 33.09 lm at 6-month FU (Relative change at 1month FU compared to baseline of 31.7%; relative change at 6month FU of 28.5%; P = <0.001, respectively). We observed a decrease in maximum expansion of skin roughness, with S max at 18.1% at 1-month FU compared to baseline; relative change at 6-month FU of 33.7% to baseline; P = <0.01, respectively). Furthermore, an overall more plane surface was calculated, with S dr : baseline score of 9.64 AE 4.06%, 4.44 AE 1.10% at 1-month FU and 4.13 AE 1.23% at 6-month FU (Figs 5 and 6).…”

“…[15][16][17] However, various studies using light and laser techniques showed promising outcome in the improvement of SD. 2,6,18 Intense pulsed light (IPL), with visible beam of highintensity light, increased dermal collagen level and led to skin tightening. [18][19][20] A broad range of laser devices has been utilized for the treatment of SD with variable outcome.…”

“…The effectiveness of IPL in treating skin wrinkles is attributed to the creation of microthermal damage in the dermis resulting in increased collagen production in a process similar to that seen in wound healing. 13,14 Pathology specimens obtained after IPL treatment show enhanced collagen and elastic fibre formation in the papillary and sub-papillary dermis, resulting in an increase in volume which in turn leads to smoothing the upper dermal layers and the increased epidermal turnover. 15,16 In an in vitro study, Wong et al 17 showed that after IPL irradiation, the number of live fibroblasts increases more than 100%.…”
