The Use of MALDI-TOF Mass Spectrometry to Analyze Commensal Oral Yeasts in Nursing Home Residents

Abstract: Matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF MS) is a rapid and accurate method to identify microorganisms in clinical laboratories. This study isolates yeast-like microorganisms in the oral washes that are collected from non-bedridden nursing home residents, using CHROMagar Candida plates, and identifies them using Bruker MALDI-TOF MS. The ribosomal DNA sequences of the isolates are then examined. Three hundred and twenty yeast isolates are isolated from the oral was… Show more

“…Cultivation is inevitable when yeasts are identified by MALDI-TOF MS, but the process only requires a minute number of cells, and limited cultivation time. The spectra were analysed using MALDI Biotyper OC version 4.1 (Bruker Daltonik GmbH) and online software MSI (Mass Spectrometry Identification platform) (8). In case of discrepancies between MALDI-TOF and biochemical testing results, we confirmed the ID by DNA-sequencing of ITS region as previously described by our scientific team (9).…”

“…All isolates were subjected to matrix-assisted laser desorption ionization-time of flight mass spectrometry (MALDI-TOF MS) for species identification using MALDI Biotyper software (version 3.1) [ 22 ]. When isolate identifications were inconsistent with the color on CHROMagar Candida agar medium or when uncommon species were reported, the sequences of the internal transcribed spacer (ITS) region and/or the D1/D2 region of ribosomal DNA were used for species identification.…”

“…Candida is a commensal yeast of the oral cavity and may be observed in about 50% of the oropharyngeal region of healthy and non-immunocompromised individuals [4]. C. albicans is the main Candida species in the oral mycobiota, followed by C. glabrata, Candida parapsilosis, Candida tropicalis, and Candida krusei [5,6].…”
