The Use of Gas‐Liquid Chromatography for the Determination of Sugars Extracted from Soils by Sulfuric Acid

Abstract: A quantitative procedure was developed for the determination of soil sugars by gas‐liquid chromatography of the acetate derivatives of the alditols obtained by reduction of the sugars. Myoinositol or quebrachitol was used as an internal standard during the production of alditol acetates and their separation by gas‐liquid chromatography.Sugars were isolated from soil hydrolysates by neutralization of the acid and precipitation of the sulfate using strontium carbonate. The filtrates were evaporated to dryness an… Show more

“…The recovery was 85 percent for mannose and between 91 and 95 percent 210 M. YOSHIDA and K. KUMADA b) Determination of neutral sugars. Based on the methods described by BORCHARDT and PIPER (2) and OADES et al (3), the hydrolysates plus internal standard (myoinositol) were transferred into a round bottom flask (100 ml). About 0.07-0.08 g of sodium borohydride was added to the flask, mixed well and allowed to react for 5 hr.…”

“…Neutral sugar analyses were carried out in 2 steps following a procedure adapted from previous work ([ Bethge et al, 1966], [Oades et al, 1970], [Modzeleski and Laurie, 1971] and [Cowie and Hedges, 1984]). The first step, which comprises a phase of soaking with 24 N H 2 SO 4 , allows the hydrolysis of both cellulose and hemicellulose(s).…”

“…La technique utilisée, préconisée par Oades et al (1970), offre l'avantage d'extraire la plus grande partie des sucres monomères tout en limitant leur dégradation. Les sucres extraits sont dosés par chromatographie en phase gazeuse après silylation .…”
