Monosodium glutamate (MSG) is a widely used food additive, and there is a trepidation that MSG plays a critical role in multiple hepatic disorders. This study was planned to investigate Graviola extract (GE) effects on hepatic and cellular alterations induced by MSG. Fifty Wistar rats were randomly allocated into five groups: control (received normal saline), Graviola (received 200 mg/kg body weight), MSG (received 2.4 gm MSG/kg, 15% of Lethal dose (LD50) of MSG), Graviola + monosodium glutamate (MSG + GE; received GE, 200 mg/kg/day and MSG 2.4 gm/kg body weight (BW) for the next four weeks), and monosodium glutamate + Graviola (received MSG only (2.4 gm/kg BW) daily for four weeks, then concomitant with Graviola (200 mg/kg BW) daily for the next four weeks. MSG and GR were administered orally for eight weeks. Our results showed that MSG caused a significant increase in oxidative stress markers malondialdehyde (MDA), reactive oxygen species (ROS), nitric oxide (NO), hydrogen peroxide (H2O2), proinflammatory cytokines interleukin 6 (IL-6) level, a tumor protein (P53), hepatic cellular damage, as well as proapoptotic markers caspase-3, and B-cell lymphoma 2 (BCL-2)-like protein 4 (Bax). A significant decrease in superoxide dismutase (SOD), catalase (CAT), glutathione S transferase (GST), reduced glutathione (GSH), and an antiapoptotic agent B-cell lymphoma 2 (BCl-2) was observed. The detected MSG effects were normalized by Graviola administration, either a prophylactic or protecting dose. Besides, Graviola reduced the expression of inducible nitric oxide synthase (iNOS) and hepatic fatty acid synthase (FAS) and led to the upregulation of the silent information regulator protein one gene expression gene (SIRT1).In conclusion, the results suggest that Gaviola’s interrelated antiapoptotic, antioxidant, and anti-inflammatory properties are potential mechanisms to enhance hepatic deficits and protect the liver. Graviola can, therefore, be considered a promising hepatoprotective supplement. Additionally, further human clinical trials are also necessary to validate the present research.

Numerous flow cytometric analyses are based on DNA content studies. We have considered firstly monoparametric cell cycle analyses, which only take DNA content into account, but are sometimes of limited interest. Then, we have presented multiparametric analyses, which can be used to improve cycle phase identification by taking simultaneously into account DNA and other cellular components, or by considering some events occurring during cell cycle. Finally, we have discussed monoparametric and multiparametric cell cycle analysis interest in various application fields, particularly in pharmacology, toxicology, tumoral pathology and higher plant system studies.

Flow cytometry in the clinical laboratory has evolved through the development of several major technological advances, Scientific achievement combined with growing database of clinical correlation studies has stimulated the use of flow cytometry as a clinical laboratory tool.The primary scientific discovery that led to the application of flow cytometry to clinical diagnosis was the hybridoma methodology of Kohler and Milstein with the resultant surge in monoclonal antibody development. Flow technology provided much more rapid and accurate evaluation of cell phenotypes and function of sw face determinants than did the standard microscopic techniques. This was a n early step in incorporating routine flow analysis into the clinical laboratory. A growing clinical interest in ploidy and proliferative activity of tumors has directed efforts to optimize DNA and RNA analysis by flow. Improvements in analysis programs are aimed at more accurately assessing the percentage of cells in S phase. Laboratory applications continue to validate tissue processing variations to maximize recovery of nuclei from all tissues, whether submitted fresh, frozen, or in paraffin blocks. The ability to detect nuclear antigens and oncogenes by monoclonal antibodies and cDNA probes will stimulate more activity in flow evaluation of malignant tissues.The future role of flow cytometry in the clinical laboratory is a continuously evolving process. The responsibilities of the laboratory are to provide accurate evaluations of specimens submitted and to assist the physician in interpretation of these results, The requirements placed on the laboratory, by clinical necessity or by economic climate, are to perform evaluations rapidly, to communicate results clearly and efficiently, and to provide the service in a cost-effective manner.Our ability to satisfy the laboratory's responsibility is directly related to education-continuing education of the laboratory and re-education of the medical community. The generous exchange of ideas and techniques at workshops and meetings, which has become characteristic of the flow cytometry scientific community, must be encouraged to continue; our role as a "service resource" must be represented at these meetings to identify clinically important flow analyses for transfer from the research laboratories to the clinical laboratory and to contribute feedback from our "real life" applications experience.The standardization of flow analyses between laboratories is essential if they are to provide a consistent clinical resource. The challenge is to achieve equivalent clinical results in each flow laboratory, regardless of size, geographical location, or economic pressure. Only in this ability will they have the credibility to educate physicians in the proper utilization of the flow resource and to improve the quality of patient care.The education of physicians who order laboratory tests occurs through several different levels of exposure.
