Damselfish ileurofibromatosis (DNF) is a neoplastic disease affecting the peripheral nervous system and chromatophores of bicolor damselfish Pornacentruspartitus on Florida (USA) reefs. This report presents the results of transmission experiments using intramuscular and intraperitoneal injection of a variety of tumor preparations to induce DNF. Neurofibromas developed within 1 yr of injection in 76% of fish injected with homogenates of spontaneously occurring tumors with earliest tumor development occurring at 5 to 6 wk post-injection. Cell-free transmission using 0.2 pm filtrates of DNF tumor homogenates was observed, demonstrating that the agent responsible for DNF is subcellular. Four out of five tumor cell lines established from neurofibromas were found to be tumorigenic when injected, while cells cultured from normal fish had no effect, indicating that the etiologic agent of DNF was present and infectious in tumor cell lines but not in cell lines or primary cultures derived from healthy fish. Serial passages of tumors were conducted through 19 passage generations in vivo. A comparison of the cumulative time to tumor distributions showed that most experiments beyond the third serial passage yielded significantly faster tumor development than was obtained by passaging from spontaneous tumors. Induced tumors exhibited more localized growth patterns and were typically more rapidly g]-owing and more rapidly lethal than spontaneous tumors. In contrast to the differences in growth patterns observed, the histological phenotype of the spontaneous, wild-type DNF lesions was indistinguishable from that of the induced tumors obtained from serial passages as well as in experiments involv~ng cell-free filtrates and cultured tumor cells. These observations indicate that the process of neoplastic transformation is relatively stable in this system, \nth little or no phenotypic drift caused by these manipulations. Naturally occurring, transmissible cancers that arise from cells of the central or peripheral nervous system or from chromatophores have not been previously identified. Thus, DNF is a unique disease and may provide an important model for investigating thls rather unique form of viral oncogenesis.

Nociceptin/orphanin FQ (N/OFQ) and the N/OFQ peptide (NOP) receptor play important roles in regulating gastrointestinal function. To assess whether the NOP receptor is implicated in the etiopathogenesis of irritable bowel syndrome (IBS), we measured the levels of NOP receptor mRNA and protein in the jejunal and colonic tissues of healthy subjects and patients with diarrhea-predominant IBS (D-IBS) and constipation-predominant IBS (C-IBS). Mucosal biopsies were obtained from the jejunum and colon of patients diagnosed with D-IBS and C-IBS by the Rome III criteria and from healthy control subjects. The expression of NOP receptor mRNA was measured quantitatively using quantitative PCR (qPCR) and NOP protein expression was assayed immunohistochemically using a rabbit monoclonal antibody to OFQ. NOP receptor mRNA was detected in the jejunum and colon of healthy subjects and was more highly expressed in the jejunum than in the colon. Expression was lower in the jejunum and colon of patients with D-IBS; however, it was similar in patients with C-IBS and healthy subjects. The numbers of OFQ-positive cells in the jejunum and colon were similar among the three groups. The NOP receptor may be involved in the regulation of intestinal movement in healthy individuals. Its involvement in the pathophysiology of IBS may depend on whether the IBS is constipation- or diarrhea-predominant.

The progress of mitomycin C (MMC) bioreduction was studied in vivo in the rat Sp 107 mammary carcinoma after intra-tumoural injection of either 100 micrograms or 1 mg. 2,7-Diaminomitosene (2,7-DM) was utilised as a primary bioreductive metabolite and 10-decarbamoyl 2,7-diaminomitosene (DC 2,7-DM) served as a secondary bioreductive metabolite, both of which were measured by high-performance liquid chromatography. 2,7-DM and DC 2,7-DM were produced rapidly, achieving close to their maximal concentrations at the earliest time point studied [5 min]. 2,7-DM was cleared rapidly from the tumour with apparent half-lives of 5 and 35 min after the low and high drug doses, respectively. DC 2,7-DM had a longer apparent half-life of 130 min at the higher dose but, as compared with 2,7-DM, was only a minor metabolite [the area under the curve (AUC) of 2,7-DM was 5.6-fold that of DC 2,7-DM]. At the lower drug dose, DC 2,7-DM was not detectable. Rapid formation and disappearance of bioreductive metabolites of MMC may account for the failure of previous studies to detect these products in vivo.
