The Use of Dried Blood Spot Sampling in the National Social Life, Health, and Aging Project

Abstract: The high cooperation rate and quality of the spots collected suggest that the collection of DBSs in population-based research is a feasible and viable alternative to venous blood draws. The relative ease of sample collection, transport, and storage are significant benefits. Care should be taken, however, when comparing results from analysis of DBS samples with those obtained from serum or plasma samples.

“…Serum equivalents therefore were calculated by using the following algorithm based on the serum-blood spot regression: serum [high sensitivity C-reactive protein (hsCRP)] = 1.38*(blood spot CRP value) -0.97. Blood spot CRP measures have been used in several epidemiologic studies (45,46,53). Observations with values above 10 mg/L indicate frank infection and were removed from statistical analysis (n = 109 from a total of 6,000 observations), whereas values below that index the extent of chronic lowgrade systemic inflammation associated with cardiovascular and metabolic risk (54).…”

“…This improvement supports the idea that if in future studies adequate training is provided, the DBS collection technique may be used as an alternative to venipuncture in an ambulant setting. [12][13][14] No guidelines are available on the percentage of spots that should fulfill a certain quality standard. As we asked the patients in this study to generate four DBS and as we wished to perform duplicate analysis (i.e., analysis on two different spots on every time point), we put forward an acceptance criterion of 50 % (i.e., there should be at least two DBS with acceptable quality:…”

“…Since a maximum of four DBS was requested, DBS cards with at least two suitable DBS were considered to have acceptable quality. [14] For the first collection period, this criterion was fulfilled by 100 % of total cards collected by patient 1, none of the cards by patient 2 (analysis of single spots was however possible on 6 out of 7 days) and 83 % of the cards by patient 3. When repeating the study (series 2), the number of well collected DBS increased, to 71 % by patient 2 and to 100 % by patient 3 (Fig.…”

“…Nevertheless, the significant linear correlation between matched serum and DBS samples for many analytes (3,20,(27)(28)(29)(30) implies that serum or plasma equivalents can be derived from DBS values (3,28,(30)(31)(32)(33) . It is on this basis that we derived a value of 4?24 to correct the DBS CRP values to serum CRP values.…”
