The Use of Agmatine Provides the New Insight in an Experimental Model of Multiple Sclerosis

Abstract: The aim of the study is to investigate the hypothesis that agmatine (AGM) enhances blood brain barrier (BBB) compounds properties in experimental autoimmune encephalomyelitis (EAE), which is an established animal model for studying multiple sclerosis (MS). Wild-type (WT) and knockout (KO) CBA/H iNOS-/- mice, 3 months old (15 ± 5 g) were used for EAE induction by myelin basic protein (MBP) dissolved in complete Freund's adjuvant (CFA). The animals were divided into control, CFA, EAE, EAE + AGM and AGM groups. A… Show more

“…Polyamines contribute to cell proliferation, hypertrophy and tissue development. In addition to its functions in other tissues, polyamines also have a variety of tissue-specific roles that affect neuronal excitability by regulating ion channels and receptors in brain tissue (9)(10)(11)(12)(13)(14). However, despite all this information, the functions of the endogenous polyamines in CNS are still unknown.…”

“…2 Its exceptional modulatory effects on a number of molecular targets have been demonstrated, 2 including those involved in neurotransmitter systems, 2,3 nitric oxide synthesis, 2,4 and polyamine metabolism, 2,4 highlighting the potential therapeutic applications of agmatine. 2,5,6 As a result, the importance of agmatine production is increasingly being recognised in the pharmaceutical industry. Currently, agmatine can be industrially produced by chemical synthesis from 1,4-butanediamine and 1,4-butylenedibromide.…”

“…Due to the widespread presence of agmatine in the peripheral and central nervous system, it is conjectured to be a "magical shotgun" -a non-selective drug with multiple targets, potentially leading to more effective treatments 11,12 . AGM function in the central nervous system includes antidepressant-like effects 13,14 , protection against schizophrenia 15 , improvement of cognitive dysfunction in Alzheimer's disease 16 , and improvement of multiple sclerosis 17 .…”
