The UL12 Protein of Herpes Simplex Virus 1 Is Regulated by Tyrosine Phosphorylation

Abstract: The herpes simplex virus 1 (HSV-1) UL12 protein (pUL12) is a nuclease that is critical for viral replication in vitro and neurovirulence in vivo. In this study, mass spectrometric analysis of pUL12 and phosphate-affinity SDS-polyacrylamide gel electrophoresis analysis identified tyrosine at pUL12 residue 371 (Tyr-371) as a pUL12 phosphorylation site: Tyr-371 is conserved in pUL12 homologs in herpesviruses in all Herpesviridae subfamilies. Replacement of Tyr-371 with phenylalanine (Y371F) in pUL12 (i) abolished… Show more

“…ICP27 is involved in a series of processes in the nucleus and the cytoplasm, where it is attributed to a number of different protein interactions. Host protein-protein interactions are often regulated by phosphorylation, and phosphorylation events are reported to regulate a number of herpesvirus proteins (39)(40)(41)(42)(43). ICP27 is observed to have both stable and transient phosphorylations (44).…”

“…We also analyzed plaque sizes in SK-N-SH cell monolayers infected with YK333(EGFP), YK491(⌬VP26/EGFP), YK492 (VP26T111A/EGFP),YK493(VP26⌬/TA-Repair/EGFP),orYK494 (VP26T111D/EGFP) under plaque assay conditions described previously (41). In agreement with the growth properties of these viruses demonstrated in this and previous studies (42), YK491(⌬VP26/EGFP) and YK492(VP26T111A/EGFP) produced plaques similar in size but that were smaller than those observed with YK333(EGFP) and YK493(VP26⌬/TA-Repair/EGFP) (Fig.…”
